Bacillus subtilis pgsE (Formerly ywtC) stimulates poly‐γ‐glutamate production in the presence of zinc

Abstract: Poly-γ-glutamate (PGA) is a versatile nylon-like material, and enhanced production of PGA is required for various bio-industrial applications. In this study, we first examined the effects of available sugars on the production of Bacillus subtilis PGA, and demonstrated the good applicability of pentoses (e.g., D-xylose). Then, we characterized the pgsE gene of B. subtilis, which encodes a 6.5-kDa protein of 55 amino acids (PgsE), as a genetic tool for increasing the yield of PGA without changing its structural … Show more

“…Bacillus species have been reported to be useful as bio-control agents against plant fungal diseases 53 . Numerous species of the Bacillus genus are reasonably considerable for the purposes, as they are functional for the production of numerous molecules and supplementary goods for foodstuff as well as in agricultural, biological and pharmaceutical industries 55,56 . In the pharmaceutical industry, there is high demand for Bacillus species because of its potential to produce a broad array of metabolites with antimicrobial activity [57][58][59][60][61] .…”

“…The role of pgsE in the production of c-PGA is dispensable since pgsB, pgsC and pgsAA at high concentrations were able to form a complex capable of producing c-PGA even in the absence of pgsE . On the contrary, Yamashiro et al (2011) found pgsE to be essential in c-PGA production as its introduction into the production medium in the presence of Zn 2+ tripled the production rate of c-PGA from B. subtilis (chungkookjang). showed that CapE (a 47 aa peptide) is also responsible and essential for the production of c-PGA as it appeared to interact with CapA.…”

“…Among these proteins, the role of PgsE remains unclear. However, we recently detected the potential Zn 2ϩ -dependent enhancer-like activity of PgsE in B. subtilis PGA synthesis (24). In this study, the secondary (viz, moonlighting; see reference 13) function of PgsE in EDM is highlighted.…”
