CalpA, a Drosophila Calpain Homolog Specifically Expressed in a Small Set of Nerve, Midgut, and Blood Cells

Abstract: Calpains are calcium-dependent proteases believed to participate in calcium-regulated signal pathways in cells. Ubiquitous calpains as well as tissue-specific calpains have been found in vertebrates. We isolated cDNA clones for a highly tissue-specific calpain gene from Drosophila melanogaster, CalpA, at 56C-D on the second chromosome. The expression of the CalpA gene product was monitored by using a specific antiserum directed against the product expressed by one cDNA clone. The encoded protein is found in a … Show more

“…Gl-CalpB combines features found in Dm-CalpA or Dm-CalpB. It has no insertion sequence in dIV, like Dm-CalpB, but has a dI length similar to that in Dm-CalpA ( Fig.·1; Emori and Saigo, 1994;Jékely and Friedrich, 1999;Theopold et al, 1995). These data suggest that Dm-CalpA and Dm-CalpB are derived from duplication of a common ancestral gene and that no such duplication occurred in crustaceans.…”

“…Atypical calpains are either truncated, e.g. lobster CalpM, nematode CPL-1, mammalian nCl-2′ and Drosophila Dm-CalpA′ (Sorimachi et al, 1993;Theopold et al, 1995;Yu and Mykles, 2003), or have the Cterminal region replaced by a different sequence, e.g. the 'T domain' in mammalian calpain 5 (Capn5) and nematode TRA-3 (Barnes and Hodgkin, 1996;Dear et al, 1997) or the 'SOL domain' in mammalian SOLH and Drosophila SOL or Calpain D (Delaney et al, 1991;Friedrich et al, 2004;Kamei et al, 1998).…”

“…In invertebrates, only a few typical calpains have been identified thus far. Drosophila melanogaster has three typical calpains, CALPA/ dm-calpain, CALPB, and CALPC [36,41,69,163]. Schistosoma mansoni and S. japonicum also have at least one typical calpain (Sm-calpain and Sj-calpain) [3,84].…”

“…In this work we corrected and extended our knowledge about calpain B, which besides calpain A described earlier [13], is the other enzymically active Ca 2+ -dependent protease in Drosophila resembling in domain structure the typical mammalian calpains. We turned our attention also to the behaviour of the enzyme in vivo, i.e.…”

“…A partial clone with sequence homology to vertebrate calpains was found serendipitously [13]; the full-length clone, coding for calpain A, was sequenced and the expression pattern of mRNA and protein in fly tissues studied. The second fly calpain homologue, calpain B, was assembled later from expressed sequence tag clones.…”
