Clusterin, a gene enriched in intestinal stem cells, is required for L1-mediated colon cancer metastasis

Abstract: Hyperactive Wnt signaling is a common feature in human colorectal cancer (CRC) cells. A central question is the identification and role of Wnt/β-catenin target genes in CRC and their relationship to genes enriched in colonic stem cells, since Lgr5+ intestinal stem cells were suggested to be the cell of CRC origin. Previously, we identified the neural immunoglobulin-like adhesion receptor L1 as a Wnt/β-catenin target gene localized in cells at the invasive front of CRC tissue and showed that L1 expression in CR… Show more

“…In this study, HBE cells were treated with 5-FU, resulting in a decrease in numbers of cells in the S phase and thus, cells in G0/G1 phase were concentrated. This phenomenon has been previously observed in several tumor cells from the gastrointestinal tract [ 25 , 26 ], lung [ 27 34 ], and liver [ 35 ]. Treatment of HBE cells with 5-FU resulted in decrease in methylation of Sox2 DNA promoter from 8.4% to 4.8%, and the methylation island disappeared.…”

“…APOJ promoted metastasis of colon cancer and promoted invasion of tumor via the p38/MAPK/MMP9 pathway . APOJ also conferred resistance of breast cancer cells to TNF α and caused apoptosis via activation of NF‐ κ B and overexpression of Bcl‐2 …”

“…The β-catenin-TCF complex was shown to regulate the energy metabolism in CRC stem cells and to fuel the growth of CRC tumors by inducing the expression of the transcription factor PROX1 78 , 79 . The cell adhesion receptor L1 (see above) induces the expression of the ISC marker Clusterin in CRC cells via STAT1 activation that is known to be stimulated by pro-inflammatory cytokines 80 . The increased expression of the ISC marker Msi1 81 , an RNA-binding protein and a β-catenin-TCF target gene, was linked to the elevated metastatic potential and poorer prognosis of CRC 82 , 83 .…”
