Csf1r-mApple Transgene Expression and Ligand Binding In Vivo Reveal Dynamics of CSF1R Expression within the Mononuclear Phagocyte System

Abstract: CSF1 is the primary growth factor controlling macrophages numbers, but whether expression of the CSF1 receptor (CSF1R) differs between discrete populations of mononuclear phagocytes remains unclear. We have generated a Csf1r-mApple transgenic fluorescent reporter mouse that, in combination with lineage tracing, Alexa Fluor 647-labeled CSF1-Fc and CSF1, and a modified ΔCsf1r-ECFP transgene that lacks a 150bp segment of the distal promoter, we have used to dissect the differentiation and CSF1-responsiveness of m… Show more

“…Wild‐type (WT) C57BL/6OlaHsd CD45.2 + , congenic C57BL/6 CD45.1 + CD45.2 + , C57BL/6OlaHsd Csf1r ‐mApple + mice, and Cdh5‐ Cre ‐ ERT2 mice crossed with the mTmG ( Rosa26 Sor tm4(ACTB‐tdTomato, EGFP)Lou /J) line, were bred and maintained in specific pathogen‐free facilities at the University of Edinburgh, UK. Mice were sex and age matched, 6–12 weeks for WT studies and 11–12 weeks for Cdh5 mice.…”

“…One possibility could be that the tissue and bone marrow environments provide signals that drive proliferation (e.g., mitogens in appropriate quantities); alternatively, monocytes within the blood could actively be inhibited from proliferating. In this regard, CSF1 is a well‐characterized Mϕ mitogen and Ly6C+ blood monocytes are able to outcompete Ly6C − monocytes for capture of circulating CSF1, 10 yet they remain nonproliferative. However, it also remains unclear whether those Ly6C + cells that proliferate within the healing skin should actually be considered monocytes.…”

“…However, formal demonstration that proliferation of monocytes is independent of CSF1 or IL‐4 is lacking. Unfortunately, it is well known that surface CD115 ( Csf1r ) is difficult to detect by flow cytometry on Mϕs isolated from tissues by enzymatic digestion, despite expression of CSF1R by these cells in vivo 10 . Furthermore, a detected loss of measurable CSF1 in tissues could reflect an elevated consumption of CSF1 that would be predicted to occur with influx of monocytes.…”
