In vitromaturation and fertilization of prepubertal and pubertal black Bengal goat oocytes

Abstract: Oocytes retrieval, in vitro maturation (IVM) and fertilization (IVF) efficiency are inevitable steps towards in vitro production of embryos. In the present study, these parameters were investigated in the ovaries of prepubertal (n = 31) and pubertal (n = 61) black Bengal goats obtained from a slaughterhouse. Nuclear maturation was evaluated upon aspiration and following IVM in TCM-199 (Earle's salt with L-glutamine and sodium bicarbonate) for 27 h at 39℃ under 5% CO2 in humidified air. The oocytes retrieval an… Show more

“…LH has been reported to increase IVM rate in oocytes collected from the ovaries of adult animals in many species (Eppig, 1996;Junk et al, 2003;Phillips and Dekel, 1982;Zuelke and Brackett, 1990). Our results are similar and in agreement with another study (Khatun et al, 2010) that also reported a similar maturation rate of 60% in black Bengal goat oocytes.…”

“…In vitro maturation rate of goat's oocytes without vitrification is similar to that reported in Bangladesh (Khatun et al, 2011) and India (Purohit et al, 2012). Contrasting to the present finding, higher rates of in vitro maturation (71.4 to 81.4%) were reported in goat oocytes when cultured without vitrification elsewhere (Izquierdo et al, 2002;Anguita et al, 2007).…”

“…These events are defined as oocyte maturation and lead to an ovulated fertilizable oocyte. For IVM, oocytes are incubated for upto 24 h (bovines) [ 23 , 24 ]; 27 h (caprines) [ 22 , 25 ] and 44 h (porcines) [ 26 ] in medium supplemented with hormones (gonadotropins) and growth factors under mineral oil at 39 °C in 5 % CO 2 in humidified air. Gonadotrophins play a major role in triggering resumption of meiosis in the oocyte and expansion of cumulus oophorus.…”
