In vitro Selection for Fusarium Wilt Resistance in Gladiolus

Abstract: Cormels pieces of four Fusarium susceptible Gladiolus cultivars (Friendship, Peter Pears, Victor Borge and Novalux) formed friable calli when cultured in vitro on Murashige and Skoog basal medium containing various concentrations of auxin and cytokinin. The friable calli established cell suspensions. Plantlet regeneration was obtained from the control callus, control cell suspension derived callus and in vitro selected Fusarium oxysporum Schlecht. resistant cell-lines of Friendship. The in vitro cormlets showe… Show more

“…Virus-eradication has been successfully reported (Simonsen and Hildebrandt 1971;Logan and Zettler 1985;Mangal et al 2002), and plantlets resistant to fusaric acid (produced by Fusarium oxysorum) were regenerated from suspension cultures by in vitro selection methods (Remotti et al 1997). Similar experiments using four different cultivars ('Friendship', Novalux', 'Peter Pears', and 'Victor Borge') yielded plantlets that displayed improved growth when inoculated with conidia or spores of F. oxysporum compared to unselected control cell lines (Nasir and Riazuddin 2008).…”

“…Previous studies have demonstrated that the albino or yellow phenotype can result from multiple factors, including Chl biosynthesis or degradation (Lin et al, 2014 ), tetrapyrrole synthesis for heme and phytochromobilin (Yaronskaya et al, 2003 ), carotenoid biosynthesis, or degradation (Qin et al, 2007 ; Wang et al, 2009 ), chloroplast development and biogenesis (Peng et al, 2012 ; Su et al, 2012 ), signal transduction during chloroplast development (Sheng et al, 2014 ), disease resistance (Nasir and Riazuddin, 2008 ), reduced light-harvesting Chl proteins (LHCPs) (Asakura et al, 2008 ), and photo-oxidative stress (Miura et al, 2010 ; Zhan et al, 2014 ). Digital gene expression (DGE) analysis of the tea cultivar Anji Baicha identified several genes involved in Chl biosynthesis or chloroplast development (Ma et al, 2012 ).…”

“…The present study revealed the production of chrysanthemu m plants with greater resistance to leaf spot disease than the lines produced by other methods [17,20,21]. The improvement in leaf spot disease resistance using this protocol over a period of four years was more than achieved earlier [16].…”
