In VivoImmunomodulation and Lipid Peroxidation Activities Contributed to Chemoprevention Effects of Fermented Mung Bean against Breast Cancer

Abstract: Mung bean has been reported to have antioxidant, cytotoxic, and immunomodulatory effects in vitro. Fermented products are reported to have enhanced immunomodulation and cancer chemopreventive effects. In this study, fermented mung bean treatments in vivo were studied by monitoring tumor development, spleen immunity, serum cytokine (interleukin 2 and interferon gamma) levels, and spleen/tumor antioxidant levels after injection with low and high risk 4T1 breast cancer cells. Pretreatment with fermented mung bean… Show more

“…Upon the experimental end-point, histopathology of 4T1 MIND tumors was examined to determine if their features differed from what has been reported for 4T1 MFP tumors. Trichrome staining revealed adjacent muscle invasion ( Figure 2A) and regions of necrosis with extensive immune cell infiltration ( Figure 2B), features that have also been identified in 4T1 MFP tumors (31,32). In addition, 4T1 MIND tumors also showed abundant collagen deposition ( Figure 2C) and regions of fibrosis ( Figure 2C), two indicators of malignant progression that have not been reported for 4T1 MFP tumors as far as we are aware (33,34).…”

“…Immunophenotyping of spleen CD3, CD4, and CD8 were done based on Yeap et al 21 Briefly, the harvested spleens from normal, untreated, and kefir water–treated groups were meshed, and red blood cells were lysed using lysis buffer (8 g NH 4 Cl, 1 g Na 2 EDTA, 0.1 g KH 2 PO 4 at pH 7.4). The cells were washed with PBS and stained with the CD3, CD4, and CD8 antibodies (Abcam, USA; Catalog No: ab11089, ab25475, ab22378) for 2 hours.…”

“…Also, a non-protein amino acid, gamma-aminobutyric acid (GABA) has been shown to demonstrate anti-inflammatory activity. In vivo experiments using GABA-rich extracts were conducted on female Balb/c mice inoculated with Yac-1 and 4T1 carcinogenic cell lines [ 34 ]. Mice supplemented with diets of either 200 mg/kg or 1000 mg/kg body weight of GABA exhibited delayed tumour formation.…”

“…This was correlated with high anti-cancer cytokine levels, splenic T cell populations, splenocyte cytotoxicity and spleen/tumour antioxidant levels. Mitotic divisions in spleen were also reduced which was accompanied by a reduction in antioxidant and nitric oxide levels [ 34 ].…”
