Lactobacillus Casei Zhang Alleviates Shrimp Tropomyosin‐Induced Food Allergy by Switching Antibody Isotypes through the NF‐κB‐Dependent Immune Tolerance

Abstract: Scope: Shellfish allergy is an important cause of food allergy, and tropomyosin (TM) is the major allergen within shellfish. Probiotics are safe bacteria that benefit host health and nutrition and is proposed as a novel approach for treating immunological diseases, including food allergies. Methods and results: The probiotic strain Lactobacillus casei Zhang (LcZ) isolated from koumiss is investigated for its capacity to modulate food allergyinduced by TM in BALB/c mice. Oral administration of LcZ attenuated al… Show more

“…Over the last 15 years, the LcZ was widely used in clinical research (or animal model) and exhibited excellent health-promoting properties compared to other L. casei isolates. For example, the LcZ could alleviate shrimp tropomyosin-induced food allergy by switching antibody isotypes through the NF-κB-dependent immune tolerance 27 ; LcZ maintained the intestinal microbiome homeostasis of the sailors and prevented sailor anxiety during a long sea voyage 28 ; the fermented milk containing LcZ alleviated constipation symptoms through regulation of intestinal microbiota, inflammation, and metabolic pathways 29 ; LcZ prevents jejunal epithelial damage to early-weaned piglets Induced by Escherichia coli K88 via regulation of intestinal mucosal integrity, tight junction proteins and immune factor expression 30 . Accordingly, we further compared the transcriptomic profile of LcZ in vivo and in vitro samples and mainly focused on differences in mRNA and sRNA expression.…”

“…There is evidence that probiotics are beneficial for gastrointestinal diseases. For example, the probiotic strain Lactobacillus casei Zhang (LCZ) isolated from koumiss can alleviate allergic intestinal epithelial injury through NF-kB pathway [37]. The oral administration of fermented milk with Bacillus subtilis (JNFE0126) significantly reduced the disease activity index (DAI) of IBD and balanced the intestinal flora by increasing the abundance of Bacillus, Alistipes, and Lactobacillus, and by decreasing the abundance of Escherichia and Bacteroides [38].…”

“…Mice were sensitized according to previous research and modified by intraperitoneal injection of 50 μg of β-LG (Sigma Aldrich Co., LTD, USA) and 100 μg of Imject™ Alum Adjuvant (Alum, Thermo Fisher Scientific Co., LTD., USA) in 200 μl of sterile PBS on day 14, and boosted with 100 μg of β-LG and 100 μg of Alum on day 28 in 200 μl of sterile PBS ( 15 ). Bacterial suspension (0.2 ml, containing 10 8 live bacteria) and PBS (0.2 ml) were administered via oral gavage to β-LG-sensitized mice from day 7 to day 52 in the KV9 groups, FN3 groups, and control groups, respectively ( 16 ). Hydrocortisone (0.5 mg/kg bw, Aladdin Reagent Co., LTD., China) in 200 μl of PBS was administered via oral gavage to β-LG-sensitized mice from day 28 to day 52 in the medicine group.…”

“…Single cells isolated from spleen and intestinal lymph node were stained for FACS analysis as described previously ( 16 ). Lymphocytes were stained using the Zombie Violet™ Fixable Viability Kit (BioLegend Biotechnology Co., LTD, China) and were stained for surface markers including CD3-APC-Cy7 and CD8-APC (Thermo Fisher Scientific Co., LTD., USA); cells were then fixed and permeabilized by the Intracellular Fixation and Permeabilization Buffer Set (Thermo Fisher Scientific Co., LTD., USA) and stained for intracellular expression markers such as IFN-γ-PerCP and IL-4-PE (Thermo Fisher Scientific Co., LTD., USA).…”
