M. tuberculosis‐Complex Specific T‐Cell Stimulation and DTH Reactions Induced With a Peptide from the 38‐kDa Protein

Abstract: An immunodominant T-cell-stimulatory epitope located near the carboxy terminus of the 38-kDa antigen from M. tuberculosis (38.G, residues 350-369) was found to be M. tuberculosis-complex specific. This was demonstrated by the presence of proliferative and delayed type hypersensitivity (DTH) responses in mice immunized with Mycobacterium tuberculosis and Mycobacterium bovis BCG, whereas mice immunized with M. avium or other non-tuberculous species of mycobacteria showed no such responses. Peptide 38.G stimulate… Show more

“…As a surrogate marker for some of the key events involved in the expression of this type of protective immunity, the DTH reaction in the skin is often investigated (27). The absence of E-and P-selectin ligands on Th1 and Tc1 lymphocytes was previously demonstrated to annul trafficking of these T cells to inflamed cutaneous sites, as demonstrated by the loss of cutaneous hypersensitivity against dinitrofluorobenzene (14).…”

“…2 In this study, we transduced the murine melanoma cell line B16-B78 with a Mycobacterium tuberculosis (strain H37Rv) gene encoding a 38-kDa protein (Ag38), 9 which is one of the most immunogenic of the Mycobacterium tuberculosis antigens. 10 Transduced cells were injected into mice and the immune response against parental B16-B78 melanoma cells was evaluated.…”

“…This indicates that the recombinant 27-kDa antigen is well recognized by the cellular immune response in mice during an experimental BGC infection. It might be of interest to test in the future whether this antigen can be used as a substitute of PPD to detect TB or contact patients or animals, as has been proposed for the 38-kDa and DPPD antigens (12,40).…”
