mig - 14 Is a Salmonella Gene That Plays a Role in Bacterial Resistance to Antimicrobial Peptides

Abstract: It was previously demonstrated that the mig-14 gene of Salmonella enterica serovar Typhimurium is necessary for bacterial proliferation in the liver and spleen of mice following intragastric inoculation and that mig-14 expression, which is induced within macrophages, is under the control of the global regulator PhoP. Here we demonstrate that the mig-14 promoter is induced by growth in minimal medium containing low magnesium or acidic pH, consistent with regulation by PhoP. In addition, mig-14 is strongly induc… Show more

“…In the final step, the 4-amino-N-phenylbenzenesufonamide product was condensed with phenylisothiocyanate in dry tetrahydrofuran in the presence of triethylamine at reflux for 48 h. After concentration, the crude oily product was purified by flash chromatography on silica gel in acetone-hexane (1:3) to deliver LED209 as a white crystalline solid (melting point of 161 to 163°C) in 25% yield. The structure and purity of the product was unequivocally confirmed by 1 H NMR, which showed the expected resonances at ␦ 7.00 to 7.47 (9H, 5 ϫ m, ArH), 7.70 (5H, s, Ph), 10 MudJ mutagenesis. MudJ mutagenesis was done as described previously (35).…”

“…These modifications reduce the inflammatory properties of lipid A (LPS) and increase ST IN bacteria resistance to CAMPs and bile salts (8,16). ST IN bacteria also modify the proteins of the outer membrane through increased expression of Mig14, VirK, and PgtE that render ST IN bacteria more resistant to CAMPs (19,20). It is unclear what other advantages these modifications may have for bacterial survival and/or evasion.…”

“…This protein is required for full virulence and is under the control of the Salmonella PhoPPhoQ system (Valdivia et al, 2000). The mig-14 gene is strongly induced by polymyxin B in a phoP-independent manner and is necessary for resistance towards polymyxin B and other CAMPs (Brodsky et al, 2002(Brodsky et al, , 2005. The P. aeruginosa PA5003 is located in the core oligosaccharide biosynthesis gene cluster, downstream of PA5004 encoding a putative glycosyltransferase WapH (Lam et al, 2004;Matewish, 2004).…”
