Mycobacterium tuberculosisESAT6 Drives the Activation and Maturation of Bone Marrow-Derived Dendritic Cells via TLR4-Mediated Signaling

Abstract: 6-kDa early secretory antigenic target (ESAT6), a virulent factor of Mycobacterium tuberculosis , is involved in immune regulation. However, the underlying mechanism behind the activation and maturation of dendritic cells (DCs) by ESAT6 remains unclear. In this study, we investigated the effect on TLRs signaling on the regulation of ESAT6-induced activation and maturation of DCs. ESAT6 induced production of IL-6, TNF-α, and IL-12p40 in bone marrow-derived dendritic cells (BMDCs) from wil… Show more

“…To understand the relevance of ESAT-6 rich EVs obtained from SN-PTB subjects, we performed two experiments: first, EVs of patient #2 and #7 containing equivalent of 1-50 ng/ml ESAT-6 was added to bone marrow derived immature dendritic cells (DC) on day-6 culture (5×10^4 mouse BM mononuclear cells treated with GM-CSF 20ng/ml for 6 days; (52), and 48 hours after, In-cell western assay (53) was performed to evaluate the activation of MHC II and CD11c, two markers for DC activation. The values of MHC II and CD11c were compared with the results obtained by using recombinant ESAT-6 protein in a 50ng-10 ug dose range.…”

“…Heat shock protein 70 kDa (Hsp70) is one of the M. tuberculosis Ags that stimulate the secretion of IL‐6, IL‐12p40, and TNF via MyD88‐ and TRIF‐dependent pathways in DCs 160 . The maturation of DCs and induction of the Th1 immune response elicited by the ESAT6 Ag was found to be dependent on TLR4 but not on TLR2 161 . The apyrase conserved region 1 (Acr1) or heat shock protein X (sHSPX) M. tuberculosis , a latency Ag, induced a tolerogenic phenotype in DCs, which was characterized by the expression of programmed death‐ligand 1 (PD‐L1), T‐cell Ig and mucin‐domain containing‐3 (Tim‐3) inhibitory molecule, IDO, and IL‐10.…”
