N-acetyl-l-cysteine improves renal medullary hypoperfusion in acute renal failure

Abstract: This study evaluated the effects of N-acetyl-L-cysteine (NAC), a free radical scavenger, and N(omega)-nitro-L-arginine methyl ester (L-NAME), a nitric oxide (NO) synthesis inhibitor, on the changes in renal function, intrarenal blood flow distribution (laser-Doppler flowmetry), and plasma peroxynitrite levels during the acute renal failure (ARF) produced by inferior vena cava occlusion (IVCO; 45 min) in anesthetized rats. Renal blood flow fell on reperfusion (whole kidney by -45.7%; cortex -58.7%, outer medull… Show more

“…In our work,NAC preincubation reduces ONOO À anion release without affecting O À 2 anion production after an in vitro overnight treatment of SSc macrophages. These data are in agreement with early reports showing similar NAC effects on different cell types [23][24][25] and in vivo [26].…”

“…The formation of these nitrothiol compounds can preserve and accumulate NO in a biologically active form. A similar mechanism could explain the NAC protective effect described by Conesa and coauthors in their acute renal failure in vivo model [26] in which the presence of NO is essential for attempting NAC beneficial effect. On the other hand, NAC is unable to modify the O À 2 production, ruling out a scavenging effect on this ROS.…”

“…61 Also, Conesaet al found that NAC reduced renal vasoconstriction and improved renal blood flow. 62 The observed increases in SOD, CAT and GSH levels in NAC and MT pretreated rats could be attributed to the stimulatory synthesis and regeneration of these endogenous antioxidants by NAC and MT. MT has been reported to stimulate the activities of endogenous antioxidants and increase gene expression that improves the total antioxidant defense capacity of organisms.…”
