N‐Glycosylation is essential for the secretion of extracellular superoxide dismutase

Abstract: Extracellular superoxide dismutase (EC-SOD or SOD3) protects against various oxidative stress-related diseases by scavenging reactive superoxides in the extracellular space. It is the only SOD isozyme that is secreted and glycosylated (at asparagine 89). However, the physiological roles of its glycosylation are poorly understood. In this study, we found that the glycosylation site on EC-SOD is well conserved and that a glycosylation-deficient EC-SOD mutant retains its enzymatic activity, but is not secreted. T… Show more

“…N ‐Glycosylation of the OST subunit ribophorin (DEIGNISTSNVR) as well as calreticulin (EDELTHVYTLIISANNTFK) and a vacuolar sorting receptor (NITIPAAFIK) is in line with reports from other organism (Denning et al ., ; Sanjay et al ., ; Shen et al ., ). Superoxide dismutase (NQAGGDFNHIIYFSTLKPNGSQPSER) and vanadium‐dependent haloperoxidase (DFNITNQISAR) are both connected to hydrogen peroxide with N ‐glycosylation of the former being described as essential for its secretion (Ota et al ., ). Mannan endo‐1,4‐beta‐mannosidases (WFNQTYDQLR) catalyze the hydrolysis of mannan, a key polymer in hemicelluloses, and were shown to be N ‐glycosylated when expressed in Pichia pastoris (Do et al ., ).…”

“…Changes in the glycosylation pattern of Apa lead to a reduced stimulatory T-lymphocyte response, exhibiting the biological role of the glycan moiety 50 . Glycosylation is also required for proper localization of superoxide dismutases (SodB) 51 . The immunogenic glycoproteins MPT64 and Apa are virulence factors involved in Mtb infection of human cells and is a promising candidate for a subunit-based anti-TB vaccine 12,52 .…”

“…Immature EcSOD protein has an N-terminus with targeting signal that instructs its secretion to the extracellular space. Once this targeting signal is cleaved, the mature form gains enhanced protein stability due to a glycosylation of asparagine at position 89 (80).…”
