Pediococcus pentosaceus PP04 Ameliorates High-Fat Diet-Induced Hyperlipidemia by Regulating Lipid Metabolism in C57BL/6N Mice

Abstract: In this study, Pediococcus pentococcus PP04 isolated from the Northeast pickled cabbage had good gastrointestinal tolerance and can colonize in the intestine stably. C57BL/6N mice were fed a high-fat diet to build animal models and treated with Pediococcus pentosaceus PP04 to evaluate the antihyperlipidemia effect. After 8 weeks, the indicators of hyperlipidemia, liver injury, and inflammation were measured. The treatment of P. pentosaceus PP04 reduced the gain of total cholesterol (TC), triglycerides (TG), lo… Show more

“…P. pentosaceus has long been engineered and applied as bacterial factory to produce valuable substances ( Wu et al, 2004 ; Sun et al, 2019 ). More and more recent studies have illustrated that P. pentosaceus could exert comprehensive host beneficial effects, e.g., antioxidant activities ( Zhang et al, 2020 ), hyperlipidemia relief ( Wang et al, 2020 ), and gut microbiota restoration ( Hao et al, 2021 ). Moreover, the combination between beneficial bacteria (probiotic) and fermentable non-digestible dietary carbohydrate (prebiotic) could manifest synergistic health improvement effects, thus has been attracted much attention in novel functional food manufacturing ( Swanson et al, 2020 ).…”

“…Huang et al, (2020) found that P. pentosaceus B49 alleviated oxidative stress in constipated mice by reducing serum malondialdehyde (MDA) levels. Wang et al (2020) found P. pentosaceus PP04 effectively improved oxidative stress induced by high-fat diet by activating Nrf2/CYP2E1 signaling pathway. The signal pathway can be enhanced superoxide dismutase (SOD) and glutathione peroxidase (GSH-px) and antioxidant activity.…”

“…of the mouse fed LAB (live bacteria or cell extracts), including superoxide dismutase (SOD) activity, glutathione peroxidase (GSH-Px) activity, malondialdehyde (MDA) content, protein amino-compound content, serum endotoxin, structural changes in gut microbial community, were measured and compared with those from the murine oxidative damage model. The significant difference was taken as a reference to evaluate the ability of LAB to intervene oxidative stress in vivo ( Wang et al, 2020 ).…”

“…The method of Wang et al ( 28 ) was used to determine autoaggregation, with some modifications. In brief, the isolated strains were cultured in MRS broth at 37°C for 18–24 h (10 9 CFU/mL).…”
