TaMCA4, a Novel Wheat Metacaspase Gene Functions in Programmed Cell Death Induced by the Fungal Pathogen Puccinia striiformis f. sp. tritici

Abstract: Programmed cell death (PCD) is a physiological process to remove redundant or harmful cells, for the development of multicellular organisms, or for restricting the spread of pathogens (hypersensitive response). Metacaspases are cysteine-dependent proteases which play an essential role in PCD. Triticum aestivum metacaspase 4 (TaMCA4) is a type II metacaspase gene cloned from 'Suwon11' wheat, with typical structural features such as peptidase C14 caspase domain and a long linker sequence between the two subunits… Show more

“…The virus-induced gene silencing (VIGS) system is an effective reverse genetic tool in barley and wheat (Lee et al , 2012; Wang et al , 2012; Tang et al , 2013). In this study, the plasmids used for virus-induced gene silencing were constructed as described by Holzberg et al , 2002.…”

“…During the bombardment, the leaves were tightly placed in dishes and shot with a Bio-Rad He/1000 single-barrelled particle delivery system according to a protocol described previously (Schweizer et al , 2000; Douchkov et al , 2005; Wang et al , 2012 a ). All of the plasmids were prepared at approximately 1 μg μl –1 .…”

“…PCD has been also studied in the budding yeast Saccharomyces cerevisiae, leading to the discovery of metacaspases in execution of PCD (Madeo et al, 2009). Lately, metacaspases have been also shown to play an important function in the execution of PCD in Arabidopsis thaliana [91,92].…”
