Trifolium pratense L. (red clover) extract and doxorubicin synergistically inhibits proliferation of 4T1 breast cancer in tumor‐bearing BALB/c mice through modulation of apoptosis and increase antioxidant and anti‐inflammatory related pathways

Abstract: Therapeutic strategies against triple‐negative breast cancer (TNBC) are associated with drug‐induced toxicities. The tropical edible red clover (Trifolium pratense L.) is rich in polyphenolic compounds which confer the plant potential anticancer properties. The aim of this study was to investigate the effects of T. pratense and doxorubicin (DOX) on the apoptosis and proliferation of 4T1 tumor cells in an allograft model of tumor‐bearing BALB/c mice. Fifty‐six female 4T1‐tumor bearing‐ BALB/c mice were randomly… Show more

“…Our study showed that IL-12 was increased in E0771 tumors by treatment with combination IMT and PTX, potentially stimulating local production of IFN-γ in the TME [51]. These results are important as other studies confirm increased apoptosis and chemotherapeutic treatment response in 4T1 cells through stimulation of T cell cytokine production, namely, IL-12 and IFN-γ [51][52][53].…”

“…The animal treatment protocol was approved by the Italian Ministry of Health (authorization number 807/2017-PR). When the tumor reached volumes of around 150-200 mm 3 , the animals were treated three times at 1-day intervals (days 0, 2 and 4) intravenously via the tail vein at a dose of 5 mg/kg of doxorubicin (26)(27)(28).…”

“…All the treatments continued for 35 days. Trifolium pratense extract and DOX were dissolved in 0.5 ml DW (Akbaribazm et al, 2020a(Akbaribazm et al, , 2020b; El-Gendy, 2012).…”

“…The plant extract has been described to improve pneumonia, meningitis, skin inflammation, and some reproductive and neuronal disorders (Booth et al., 2006). In our previous study on flavonoid and isoflavone compounds by high‐performance liquid chromatography–electrospray ionization–mass spectrometry (LC‐ESI‐MS), we showed that the leaves of this plant contained formononetin, genistein, biochanin A‐7‐glucoside, daidzein 7‐O‐β‐D glucoside, and biochanin A isoflavones, as well as 3‐O‐(Z)‐p‐coumaroylquinic acid, epigallocatechin, caffeic acid, quercetin‐3,7‐diglucoside, apigenin‐7‐O‐glucoside gallocatechin, and apigenin 6,8‐diglucoside flavonoids (Akbaribazm, Khazaei, & Khazaei, 2020a, 2020b).…”
