Tu-Teng-Cao Extract Alleviates Monosodium Urate‐Induced Acute Gouty Arthritis in Rats by Inhibiting Uric Acid and Inflammation

Abstract: Gouty arthritis is an inflammatory joint disease closely related to hyperuricemia. It is characterized by deposition of monosodium urate crystals in the joints, resulting in an intense inflammatory process and pain. Control of hyperuricemia and anti-inflammation treatments are the main therapeutic approaches. However, the commonly used drugs for inhibiting uric acid and acute gouty arthritis have obvious gastrointestinal and renal toxicity; thus, there is an urgency to develop new alternative therapeutic drugs… Show more

“…Gouty arthritis is characterised by an intense inflammatory process and pain caused by the deposition of MSU crystals within the joints and connective tissues [ 21 ]. MSU crystals activate the secretion of inflammatory cytokines, including IL-1β, TNF-α, and IL-6 by macrophages, neutrophils, and monocytes-macrophages [ 22 ].…”

“…MSU crystals are one of the most effective proinflammatory stimuli, through their ability to trigger, amplify, and sustain a strong inflammatory reaction in the joint cavity [ 29 ]. MSU crystals stimulate the synthesis and release of IL-1β, a key inflammatory cytokine that regulates the differentiation, proliferation, and apoptosis of cells in gout arthritis [ 30 ]. In addition, IL-1β induces the expression of a wide range of cytokines, including TNF-α and IL-6, resulting in a large influx of neutrophils into the synovium [ 31 ].…”

“…The high (3.0 g/kg/d)/(2.0 g/kg/d), medium (1.5 g/kg/d)/(1.0 g/kg/d), and low (0.75 g/kg/d)/(0.5 g/kg/d) doses used in this study were converted based on the adult dose [ 29 ], which were equivalent to twice, equal, and half of the human dose. The positive control drugs used in this study were benzbromarone (BBR) (10 mg/kg/d, 20 mL/kg) and colchicine (COL) (0.3 mg/kg/d, 10 mL/kg), which were equivalent to the adult dose [ 17 , 30 , 31 ].…”

“…Herbal drugs have been used to treat gout in Traditional Chinese Medicine for more than 2000 years [ 17 ]. Many studies have been discovering active antigout agents derived from traditional herbal medicines [ 18 20 ].…”
