Total polyphenol content (TPC), total flavonoid content (TFC), total anthocyanin content (TAC), and proanthocyanidin (PAC) content were determined in fruit from three tropical Vaccinium species (Vaccinium consanguineum, Vaccinium floribundum, and Vaccinium poasanum) from Costa Rica sampled at three stages of fruit development. Results show that TAC increased as the fruit developed, while TPC, TFC, and PAC content decreased. Anthocyanin profiles were evaluated using electrospray ionization tandem mass spectrometry. Cyanidin and delphinidin glycosides were the predominant anthocyanins for the three tropical Vaccinium species. Proanthocyanidins were characterized using attenuated total reflection Fourier transform infrared spectroscopy, nuclear magnetic resonance, and matrix-assisted laser desorption/ionization time-offlight mass spectrometry. The presence of procyanidin structures with B-type interflavan bonds were observed, but deconvolution of mass spectrometry isotope patterns indicated that PACs with one or more A-type interflavan bonds accounted for more than 74% of the oligomers at each degree of polymerization. PAC structures vary by the nature of the monomer unit, interflavan linkage, and degree of polymerization (DP). 13 PACs are classified by the type of flavan-3-ol monomers, such as properlagonidins, which are oligomers of (epi)afzelechin, procyanidins, which are oligomers of (epi)catechin, and prodelphinidins, which are oligomers of (epi)gallocatechin. 14 However, many types of PACs contain more than one type of monomeric flavan-3-ols, such as PACs in grape seeds. PACs can have B-and A-type bonds. B-type bonds are defined by the presence of C 4 −C 8 or C 4 −C 6 interflavan bonds, whereas Atype bonds have C 4 −C 8 bonds but also have an additional interflavan bond between C 2 −O−C 7 or C 2 −O−C 5 . 9,13,15

The genus Vaccinium contains about 400 species distributed worldwide, but only a few species and varieties have had their phenolic composition and biological activity documented. In this study, phenols, flavonoids, anthocyanins, antioxidant and antimicrobial activity of methanolic extracts of Vaccinium stenophyllum Steud. fruits: 1-totally immature, 2-immature, 3-immature/close to maturity and 4-mature, were determined using UV-Visible spectrometry and HPTLC. The totally immature fruit extract showed the highest content of total phenols (19.153 ± 0.175 mg GAE/g DW), chlorogenic acid (20.867 ± 0.240 mg CAE/g DW), and the highest antioxidant activity by ABTS●+ (196.761 ± 0.641 µM TE/g DW) and DPPH● (146.580 ± 6.466 µM TE/g DW). Immature, immature/close to maturity and mature fruits extracts, exhibited the lowest MIC (9.37 mg/mL) and MBC (18.75 mg/mL) against Escherichia coli, Salmonella choleraesuis, and Shigella flexneri. The mature fruits extract exhibited the highest content of total anthocyanins (0.141 ± 0.004 mg CE/g DW) and cyanidin-3-glucoside (19.230 ± 0.309 mg CGE/g DW). The content of phenols, flavonoids and anthocyanins was higher than that reported for other Vaccinium species. These results showed the relevance of Vaccinium stenophyllum Steud. for breeding purposes to enhance the bioactivity of cultivars, or as a source of natural additives for the food industry, among others.

Vaccinium consanguinium es un arándano nativo de Costa Rica que presenta cualidades nutricionales y antioxidantes que lo hace un fruto de alto valor medicinal y nutricional. Su adaptación a las zonas altas del país lo posiciona como especie de interés, por lo que, disponer de técnicas para la obtención de material de siembra clonal es recomendable. El objetivo de esta investigación fue continuar con el desarrollo del protocolo para la micropropagación de esta especie. Se validó la etapa de establecimiento in vitro y se incluyó el uso de desinfectantes más efectivos como la combinación de jabón enzimático (5 ml.l-1) y hipoclorito de sodio (3% i.a.) en esta etapa. Se logró establecer material de campo en el invernadero mediante la siembra de plantas, enraizamiento de estacas y germinación de semillas sin necesidad de usar ningún tipo de reguladores de crecimiento y se observó 100% de supervivencia y adaptación a las condiciones ambientales del invernadero. Una señal de la adaptación mostrada por las plantas recolectadas en el campo fue que el 40% presentó floración y fructificación. Con las estacas establecidas asépticamente se evaluó el efecto de tres reguladores del crecimiento en el medio de cultivo WPM al 50%, BA (0, 3, 5, 10, 15 y 20 mgl-1), Z (0, 0,1, 0.5 mgl-1), observándose que ambas citocininas fueron efectivas en concentraciones de 3 mgl-1 y 0,1 mgl-1 respectivamente , pero el mejor procedimiento para la brotación y multiplicación consistió en la exposición de las microestacas a un choque térmico a 5ºC por 15 días, utilizando el mismo medio con 120 gl-1 de sacarosa y 0,1 mgl-1 de Z. Posteriormente, los materiales fueron transferidos al medio WPM líquido suplementado con 30 gl-1 de sacarosa y 0,1 mgl-1 de Z con puente de papel de filtro, lo que disminuyó la oxidación de los explantes y mejoró las condiciones nutricionales del cultivo reflejándose en mayor crecimiento y mejor apariencia de los brotes.
