Identification and Characterization of a Cis-Encoded Antisense RNA Associated with the Replication Process of Salmonella enterica Serovar Typhi

Abstract: Antisense RNAs that originate from the complementary strand of protein coding genes are involved in the regulation of gene expression in all domains of life. In bacteria, some of these antisense RNAs are transcriptional noise whiles others play a vital role to adapt the cell to changing environmental conditions. By deep sequencing analysis of transcriptome of Salmonella enterica serovar Typhi, a partial RNA sequence encoded in-cis to the dnaA gene was revealed. Northern blot and RACE analysis confirmed the tra… Show more

“…coli , and asDnaA in Salmonella enterica . DicF inhibits translation of the cell-division protein FtsZ when overexpressed [ 62 ], while asdnaA is expressed in stationary phase and under other stress conditions and seems to increase stability of the dnaA mRNA by an unknown mechanism [ 63 ]. A few sRNAs have been reported to be involved in bacterial cell differentiation processes that may include modulation of cell cycle control.…”

“…For instance, one of the island-encoded sRNAs, IsrM, is important in the invasion by Salmonella of nonphagocytic cells [29]. Recently, we applied the RNA-seq technology to a transcriptomic analysis of S. Typhi, and found large numbers of putative ncRNAs, some of which were identified [15,30].…”

“…With an RNA-seq analysis of the transcriptome of S. Typhi under different environmental stresses, combined with bioinformatic tools, we have recently identified many putative novel ncRNAs in bacteria [15]. A 236-nt transcript partially encoded by the minus strand of bax was expressed and designated 'Asb' (antisense RNA of bax).…”

“…The diversity of the cis-encoded ncRNA and their regulatory roles vary according to organisms as. For example, Salmonella enteric serovar Typhimurium possesses the cis-encoded ncRNA lesR-1 whose function is to control replication in eukaryotic cells [51] and Salmonella serovar Typi possesses the cis-encoded ncRNA AmgR involved in virulence in rats [52] and AsdA which regulates intracellular replication [53].…”
