Identification and mapping of human papillomavirus type 1 RNA transcripts recovered from plantar warts and infected epithelial cell cultures

Abstract: Multiple spliced transcripts of human papillomavirus type 1 were detected by electron microscopic analysis of R-loops formed with total RNA extracted from plantar warts and with poly(A)+ RNA isolated from cultured keratinocytes infected with human papillomavirus type 1. The 5' ends of the RNAs were mapped to sites in the E7 open reading frame (ORF), just upstream of the E6 ORF and in the upstream regulatory region. Species with 5' ends in E7 accounted for over 95% of all transcripts seen. Two polyadenylation s… Show more

“…This protein is a nonstructural late protein expressed to very high levels in HPV-1induced papillomas (9) and causes the disaggregation of the cytoplasmic skeleton (10). Furthermore, in HPV-1, -6, or -11 condyloma and in low-grade lesions of HPV-16, and E4 mRNA is the most abundant transcript (5,6,8,30), and antibodies to E4 of HPV-8 (33) and of HPV-16 (20) were detected in human sera. The situation with CRPV is somewhat different.…”

“…1). E1^E4 mRNA species have been demonstrated to be very abundant in HPV-1-, HPV-6-, and HPV-11-induced condylomas (8,9,24,25), as well as in an HPV-16-transformed rodent cell line (36,37). In addition, an HPV-11-specific E1^E4 transcript was successfully used as a direct marker for HPV-11 infection in both the xenograft system and in cultured human cells (4,33,34).…”

“…The upstream regulatory region (URR) contains elements involved in the control of virus transcription and replication (see text for references). (31)(32)(33) suggesting that they can represent but only a small fraction of the papillomavirus transcription products. The present communication reports the discovery of a set of mRNAs from the El gene region of BPV-1 which have not previously been described.…”
