Identification and molecular characterization of begomovirus and associated satellite DNA molecules infecting Cyamopsis tetragonoloba

Abstract: Monopartite begomoviruses comprise DNA-A as the main genome and associated satellite DNAs. Viral DNA extracted from guar (Cyamopsis tetragonoloba) showing leaf curl symptoms exhibited positive amplification of coat protein (CP) gene of DNA-A component, suggesting the presence of begomovirus. Full length DNA-A was amplified by primer pair re-designed from CP gene nucleotide sequence. The associated alphasatellite and betasatellite DNA molecules were amplified and sequenced, confirming the presence of monopartit… Show more

“…Although alphasatellites are capable of autonomous replication in permissible plant cells, they depend on helper virus-encoded proteins for movement within and between plants (8). The two alphasatellites were maintained in wheat by WDIV as efficiently as by the begomoviruses (CLCuBuV and CyTLCuV) in tobacco and are thus typical of the earlier described satellites (38,46). RNA silencing is an antiviral defense mechanism.…”

“…The primer pair MF1_FOR/REV (Table 1) was used in the PCR assays to amplify the WDIV genome from wheat samples. Alphasatellites and betasatellite were amplified in PCR assays using the primer pairs nanofor/nanorev and ␤01/04 (Table 1), respectively, as described previously (38). The RCA and PCR assays were performed for all of the 460 wheat (450 symptomatic and 10 asymptomatic) samples.…”

“…The alphasatellite molecules are also associated with the begomovirus-induced diseases occurring in the Indian subcontinent [35,36]. Like the betasatellites, the alphasatellite associated with TbLCD in Pusa, Bihar, was also found to be a recombinant of major and minor parents spreading in the Indian sub-continent (Fig.…”

“…Ltd. Bangalore, India) as per the manufacturer's instruction and used as template for amplification of betasatellite DNA in PCR. Primer pair b01 (5 0 -GAAACCACTACGCTACG CAGCAGCC-3 0 ) and b04 (5 0 -ACCCTCCCAGGGGTA CACACCGCCG-3 0 ) [18] were used and PCR was performed at 94°C/5 min for one cycle as initial denaturation, 94°C/45 s, 50°C/60 s, and 72°C/90 s for 30 cycles and a final amplification 72°C/5 min. PCR yielded a DNA fragment of ca.…”
