In 2023, fungal diseases, including Botrytis cinerea (gray mold), were isolated from various strawberry fields in Korea. Ipflufenoquin, a novel class 2 dihydroorotate dehydrogenase (DHODH) inhibitor, was applied to plant pathogenic fungal species. EC50 values indicated that ipflufenoquin achieved high sensitivity in B. cinerea while maintaining broad-spectrum activity against other fungi. Fruit and greenhouse trials confirmed its robust efficacy in gray mold diseases, including those resistant to conventional fungicides like pyraclostrobin, fluxapyroxad, and benomyl. However, the ipflufenoquin application was less effective against fungal pathogens like blossom blight and Rhizopus rot, causing shifts in the fungal community and allowing less sensitive species to emerge. Structural prediction using Alphafold2 and molecular docking simulation showed ipflufenoquin was bound to the quinone binding tunnel in DHODH and highlighted the correlation between protein-ligand binding affinity and fungicide susceptibility, with species showing higher affinities being more sensitive. This study is the first to report sensitivity of strawberry fungal species to ipflufenoquin, and this fungicide is recommended in strawberry farms against gray mold disease. Additionally, we conducted in silico predictions of fungicide sensitivity across various plant pathogenic fungi using Alphafod2 and Autodock Vina. This establishes a foundation for future computational analyses to predict the effectiveness of fungicides, offering important baseline data to enhance fungicide management strategies.

Peach (Prunus persica L.) is a globally significant fruit valued for its high edible and ornamental qualities. Peach leaf spot disease has become increasingly prevalent in recent years, negatively affecting fruit quality and esthetic appeal. During the summer of 2024, symptoms of leaf spots were observed on peach trees in an orchard in Bazhong City, Sichuan Province, China. Leaf samples displaying typical spot symptoms were collected from peach orchards, and the pathogenic agents were isolated. Based on their morphological characteristics and multi-locus phylogenetic analysis, the isolated and purified fungus SCBZPP6 was identified as Nigrospora sphaerica. Furthermore, the pathogenicity of the isolated fungus was verified via Koch’s postulates. To our knowledge, this is the first report of N. sphaerica causing leaf spot on peach in China.

O mal do panamá é um dos problemas fitossanitários mais importantes da cultura no mundo. Fusarium oxysporum f. sp. cubense (FOC), agente causal induz a morte prematura das plantas, é de difícil controle e sobrevive no solo em hospedeiros alternativos por mais de 20 anos. O crescente número de espécies no complexo Fusarium tem gerado controvérsias taxonômicas para fitopatologistas. Deste modo a analise de DNA como a técnica de RAPD são ferramentas importantes para analisar o polimorfismo e estudos de filogenia para identificação de espécies, formas especiais e raças. Nosso trabalho teve como objetivo estudar a variabilidade genética de isolados de FOC de diferentes áreas geográficas, através de marcadores RAPD. A coleta dos isolados ocorreu em Alagoas, Pernambuco, Bahia, Tocantins e Honduras em bananeiras com sintomas típicos da doença. O DNA genômico foi extraído seguindo o protocolo CTAB 10% a partir de culturas monoconidiais. Nove primers foram testados que gerou 61 bandas, polimórficas que correspondeu a 8,7 por primers comprovando alta variabilidade genética. A árvore filogenética gerada pelo coeficiente de Jaccard teve 100% de suporte estatistico pelo método UPGMA que apresentou três grupos genéticos distintos A; B e C. A técnica de RAPD foi eficaz na detecção da variabilidade genética na população de FOC revelando polimorfismo. Não houve relação quanto às origens geográficas dos isolados na árvore filogenéticas gerados pelos perfiz de amplificação de RAPD. As diferenças nos clados A, B e C refletem a diversidade genética presente nas populações de FOC influenciada possivelmente por fatores geográficos, ambientais e antropogênicos.
