Identification of a Novel Methylated Gene in Nasopharyngeal Carcinoma: TTC40

Abstract: To further explore the epigenetic changes in nasopharyngeal carcinoma (NPC), methylation-sensitive arbitrarily primed PCR was performed on NPC biopsies and nontumor nasopharyngeal samples. We have shown mainly two DNA fragments that appeared to be differentially methylated in NPCs versus nontumors. The first, defined as hypermethylated, corresponds to a CpG island at the 5′-end of the tetratricopeptide repeat domain 40 (TTC40) gene, whereas the second, defined as hypo-methylated, is located on repetitive seque… Show more

“…TTC40 encodes cilia and flagella associated protein 46, which is reported to play a role in the occurrence of nasopharyngeal carcinoma and acute myeloid leukemia [ 39 , 40 ]. In our study, zebrafish with knockdown of this gene did not exhibit an abnormal phenotype in terms of LR patterning.…”

“…In all samples, adequate amounts of RNA were extracted using commercially available kits, with ≤1 ng of sample RNA/reaction required to obtain robust ddPCR results. In contrast, from 20 to 50 ng per reaction of RNA is typically required to perform RT‐qPCR from head and neck cancer cells …”
