Identification of a Scar Marker Linked to a Shattering Resistance Trait in Sesame

Abstract: Sesame (Sesamum indicum L.) is one of the most important oil crops in temperate and tropical regions and is grown worldwide over an area of 5179 (hg ha-1) to produce 5.469.024 tonnes of seed. Capsule shattering before or during harvest can cause yield losses of greater than 50%. The objectives of the present study were to evaluate the inheritance of resistance to capsule shattering in the F2 derived from a cross between Cplus1, a sesame line with shattering-resistant capsules (Sh1Sh1Sh2Sh2), and KUAOX25, a lin… Show more

“…Previously, Uzun et al (2003) crossed another non-shattering mutant line (cc3) with the Turkish variety Muganli-57, and a closely linked amplified fragment length polymorphism (AFLP) marker was identified in the resulting segregated population. Sequence-characterized amplified region markers associated with shattering resistance were later developed from F 2 lines derived from a cross between Cplus1, a sesame line with shattering-resistant capsules, and KUAOX25, a line with shattering-susceptible capsules (Phumichai et al, 2017). In the present study, we developed InDel marker related to the shattering trait, and therefore the difference between shattered and non-shattered capsule genotypes can be visualized with basic PCR systems and simple agarose gel electrophoresis.…”

“…Although heterotic improvements in performance were obtained for seed yield and yield-related traits in these studies, there was no progress with respect to suitability for mechanized harvesting. Besides conventional breeding techniques, molecular markers have also been developed for the closed capsule mutant trait for use in markerassisted selection (Uzun et al, 2003;Phumichai et al, 2017;Zhang H.Y. et al, 2018).…”

“…Seed yield reduction in sesame occurs due to high capsule shattering at harvest and uneven capsule ripening characteristics of the sesame crop ( Abdellatef et al, 2008 ; Yol & Uzun, 2019 ; Teklu et al, 2022 ). The scientific evidence indicates that capsule shattering can lead to up to a 50% yield loss during or after harvesting in sesame ( Phumichai, et al, 2017 ). Besides yield reduction, the absence of noon shattering/indehiscent cultivars suited for mechanical harvesting is another topic for sesame improvement ( Abdellatef et al, 2008 ).…”

“…Uzun et al [9] employed the Bulked segregant analysis strategy to map the closed capsule trait in an F 2 population derived from crossing the closedcapsular mutant "cc3" with the Turkish variety "Muganli-57"; a total of 72 closed-capsular linked primers were screened and a tightly linked AFLP tag was identi ed. More recently, Phumichai et al [12] constructed an F 2 population, and 192 AFLP primers were screened, of which 9 polymorphic AFLP bands were identi ed. One AFLP molecular marker, Si-SR-32-19, can differentiate between the genotype of a capsule and a closed capsule.…”
