Identification of a σ B -Dependent Small Noncoding RNA in Listeria monocytogenes

Abstract: In Listeria monocytogenes, the alternative sigma factor B plays important roles in stress tolerance and virulence. Here, we present the identification of SbrA, a novel small noncoding RNA that is produced in a B -dependent manner. This finding adds the B regulon to the growing list of stress-induced regulatory circuits that include small noncoding RNAs.

“…Specifically, we showed that the prfA* ⌬sigB strain had higher hemolytic activity (232 hemolytic units [HU]) than a prfA* strain (213.3 HU) ( Table 3). Low hemolytic activities were observed for the ⌬prfA and ⌬prfA ⌬sigB strains, consistent with the PrfA-dependent nature of hly transcription (6,54,57,71).…”

“…This hypothesis is supported by the fact that the L. monocytogenes B regulon includes Ͼ140 genes, including those encoding proteins with importance in stress response, virulence, transcriptional regulation, metabolism, and transport (59). B also transcribes genes that encode other regulators, including small RNAs (12,54) and proteins that affect RNA stability (e.g., Hfq) (13).…”

“…They have been recognized to be important regulators in biological processes in eukaryotes (60) and bacteria (61,62). So far, a large repertoire of ncRNAs has been described in L. monocytogenes, and evidence suggests that they are central to environmental adaptation (31,(35)(36)(37)(38)(63)(64)(65). As in other bacterial models, this noncoding part of the genome is an important contributor to the regulation of virulence in L. monocytogenes (35)(36)(37)(38)(39).…”

“…MicA and RybB regulating outer membrane proteins are dependent on the alternative sigma factor E (20,32,44,46) associated with membrane stress. Recently, a novel Listeria sRNA that is under the control of the stress sigma factor B was found (31). Here, we report the properties of a novel sRNA, SR2 (30), identified by our group using the same approach that was used previously for SR1 (24).…”
