Identification of an organelle-specific myosin V receptor

Abstract: Class V myosins are widely distributed among diverse organisms and move cargo along actin filaments. Some myosin Vs move multiple types of cargo, where the timing of movement and the destinations of selected cargoes are unique. Here, we report the discovery of an organelle-specific myosin V receptor. Vac17p, a novel protein, is a component of the vacuole-specific receptor for Myo2p, a Saccharomyces cerevisiae myosin V. Vac17p interacts with the Myo2p cargo-binding domain, but not with vacuole inheritance-defec… Show more

“…For each of its functions, Vac8 interacts with specific proteins (Pan and Goldfarb, 1998;Wang et al, 1998;Scott et al, 2000;Veit et al, 2001;Ishikawa et al, 2003). Therefore, palmitoylation of Vac8 could directly affect these interactions, either by forming direct lipid-protein interactions or by inducing conformational changes that permit certain protein-protein interactions.…”

“…74 For inheritance, it binds via the adaptor protein Vac17 and the myosin Myo2 to actin. 204,205 It also interacts with the ER protein Nvj1 to establish contact zones between the ER and vacuole, 206 and it is part of a complex of phosphoproteins involved in the Cvt pathway and autophagy. 21 On isolated vacuoles, Vac8 becomes palmitoylated prior to the ATP-dependent priming reaction.…”

“…Vacuoles were labeled in vivo with N-(3-triethelammoniumpropyl)-4-(6 (4-(diethylamino) phenyl) hexatrienyl) pyridinium dibromide (FM4-64; Molecular Probes, Eugene, OR) essentially as described (Ishikawa et al, 2003). In brief, a 2 mg/ml stock solution of FM4-64 in DMSO was added to early log phase cultures for a final concentration of 80 M. After 1 h of labeling, cells were washed and were then chased in fresh liquid medium for 3-4 h.…”

“…For example, vacuole inheritance utilizes the actin cytoskeleton and the class-V myosin complex, composed of Myo2p, Vac17p, and Vac8p (Hill et al, 1996;Wang et al, 1998;Ishikawa et al, 2003;Tang et al, 2003). Myo2p is one of two class-V myosin motors in budding yeast.…”
