Identification of centrosomal proteins in a human lymphoblastic cell line.

Abstract: Highly enriched preparations of centrosomes from human T‐lymphoblasts KE 37 were analyzed for their protein content. The specific pattern of polypeptides was characterized by an abundant subset of high mol. wt proteins and a major group of proteins with mol. wt ranging from 50 to 65 kd. Several immunoreactive proteins were identified, using a rabbit serum spontaneously reacting with human centrosomes. They include a family of high mol. wt ranging from 180 to 250 kd, a 130‐kd protein and a 60‐65 kd doublet. The… Show more

“…All RACE products were cloned into pBluescript-T (21). Further human sequence information was obtained from the total sequencing of an expressed sequence-tagged cDNA from the human promyeloblast cell line KG1a, PMY2245 (22). Finally, biotinylated recombinant R II was used to screen a human tracheal cDNA library in LambdaZap using a standard protein overlay technique.…”

“…The characteristics of the 350-kDa AKAP that we have cloned appear similar to those of the centrosomal AKAP350 identified by Keryer et al (9) in an R II blot overlay of isolated centrosome proteins from the KE37 human lymphoblast cell line. These investigators immunoprecipitated the protein using the serum 0013 polyclonal antibody, previously reported as a specific marker for human centrosomes (20,22,38). No further characterization of this protein in the literature has been submitted, however, since the initial report.…”

“…Mitotic centrosomes were purified from HeLa S3 cells arrested in mitosis using 10 M S-trityl-l-cysteine using the Bornens protocol (Gosti-Testu et al, 1986). Mitotic centrosomes fractionate differently than the centrosomes normally purified from asynchronous populations of cells so care must be taken to assay each fraction for centrosomes enrichment.…”

“…An increase in centrosome-specific staining with anticentrosome antibodies occurs during G2 and a dramatic decrease takes place at the onset of anaphase (Gosti-Testu et al, 1986). These centrosomal modifications, which are likely to correspond to a rise and fall of microtubule nucleating activity (for a review, see Bornens and Karsenti, 1984), may well involve changes in phosphorylation.…”
