Identification of commercial hake and grenadier species by proteomic analysis of the parvalbumin fraction

Abstract: Analysis of parvalbumin fractions through proteomic methodologies allowed the differential classification of ten commercial, closely related species of the family Merlucciidae. Muscle extracts from nine hake species of the genus Merluccius including two subspecies of Merluccius australis (australis and polylepsis) and one grenadier species Macruronus novaezelandiae with two populations (novaezelandiae and magellanicus) were evaluated by 2-DE and MALDI-TOF MS. 2-DE demonstrated that the species tested displayed… Show more

“…Analytical methods for the detection of fish in foods are necessary to monitor hygiene practices and to ensure compliance with legislation (Poms et al, 2004). A number of different techniques for the identification of parvalbumin by chromatography, isoelectric focussing, or mass spectrometry have been developed (Ross et al, 1998;Etienne et al, 2000;Carrera et al, 2006).…”

“…A peptide should be unique for precursor proteins. Parvalbumins or their fragments are recommended as unique markers of species--specifi c fi sh allergens due to high sequence variability (8,12). A single peptide marker capable of detecting several allergens can also be identifi ed (13).…”

“…The database may be suitable for the determination of epitope profi les for potential allergens belonging to families grouping proteins with amino acid sequences containing many common fragments such as caseins [Erhardt et al, 2016], tropomyosins [Marti et al, 2007] or prolamins [Minkiewicz et al, 2015]. For protein families with highly variable sequences such as fi sh parvalbumins [Carrera et al, 2006], the analysis of common epitopes in silico using the BIOPEP database may provide no results.…”
