Identification of Dietary Bioflavonoids as Potential Inhibitors against KRAS G12D Mutant—Novel Insights from Computer-Aided Drug Discovery

Abstract: The KRAS G12D mutation is very frequent in many cancers, such as pancreatic, colon and lung, and has remained undruggable for the past three decades, due to its smooth surface and lack of suitable pockets. Recent small pieces of evidence suggest that targeting the switch I/II of KRAS G12D mutant could be an efficient strategy. Therefore, in the present study, we targeted the switch I (residues 25–40) and switch II (residues 57–76) regions of KRAS G12D with dietary bioflavonoids in comparison with the reference… Show more

“…Cancer has become a global health burden with constantly increasing incidence and mortality rates every year worldwide, and the quest for potent therapeutics development is still on in the era of precision medicine ( Girisa et al, 2021 ; Ramalingam and Arumugam, 2023a ; Ramalingam and Arumugam, 2023b ; Ramalingam et al, 2023a ; Ramalingam and Arumugam, 2023b ). Alterations/modification in the function of genes/proteins triggers the oncogenic signalling pathways rather than the normal physiological conditions, and thus alterations induce cancer progression and tumorigenesis ( Tarrado-Castellarnau et al, 2016 ).…”

“…Instead of looking for new antibiotics development which consumes time and requires huge amount of funds, the existing antibiotics can be delivered specially to the site of infection with controlled release manner [ 19 ]. Aquasomes are 3-layered vesicular nano carriers used to specially delivery the drugs to the site of target which has been used in various diseases such as cancer, cardiovascular diseases and infections [ 20 , 21 ]. In general, the aquasomes consists of ceramic core which coated with polyhydroxy compound to which the drug molecule will be loaded [ 22 ].…”
