Shikonin (SKN), a highly liposoluble naphthoquinone pigment isolated from the roots of Lithospermum erythrorhizon, is known to exert antibacterial, wound-healing, anti-inflammatory, antithrombotic, and antitumor effects. The aim of this study was to examine SKN antibacterial activity against methicillin-resistant Staphylococcus aureus (MRSA). The SKN was analyzed in combination with membrane-permeabilizing agents Tris and Triton X-100, ATPase inhibitors sodium azide and N,N′-dicyclohexylcarbodiimide, and S. aureus-derived peptidoglycan; the effects on MRSA viability were evaluated by the broth microdilution method, time-kill test, and transmission electron microscopy. Addition of membrane-permeabilizing agents or ATPase inhibitors together with a low dose of SKN potentiated SKN anti-MRSA activity, as evidenced by the reduction of MRSA cell density by 75% compared to that observed when SKN was used alone; in contrast, addition of peptidoglycan blocked the antibacterial activity of SKN. The results indicate that the anti-MRSA effect of SKN is associated with its affinity to peptidoglycan, the permeability of the cytoplasmic membrane, and the activity of ATP-binding cassette (ABC) transporters. This study revealed the potential of SKN as an effective natural antibiotic and of its possible use to substantially reduce the use of existing antibiotic may also be important for understanding the mechanism underlying the antibacterial activity of natural compounds.

Effect of Irrigation of Sulfur Solution before Sowing on Growth and Root Rot Disease of Seedling in Ginseng Nursery

: To control the disease of root rot in ginseng nursery, inorganic sulfur solution of 0.1%, 1.0%, and 2.0% were irrigated by amount of 10 ℓ per 3.3 ㎡ before sowing. On the last ten days of July, Fusarium solani and F. oxysporum were similarly detected by 44.8% and 43.8%, respectively, while Cylindrocarpon destructans was low detected by 4.4% in the diseased seedling. The more sulfur's concentration was increased, the more soil pH was decreased. Soil pH was decreased from 5.87 to 4.59 by the irrigation of sulfur solution of 1.0%. The more sulfur's concentration was increased, the more electrical conductivity (EC) of soil was increased. EC was increased from 0.27 dS/m to 1.28 dS/m by the irrigation of sulfur solution of 1.0%. Irrigation of sulfur solution was effective on the inhibition of damping-off caused by Rhizoctonia solani in ginseng seedling. Control value for damping-off by the irrigation of sulfur solution of 1.0% and 2.0% were 75.7%, and 78.5%, respectively. Growth of leaf was inhibited by the irrigation of sulfur solution of 2.0%. Root weight per 3.3 ㎡ showed the peak in sulfur solution of 1.0%, while survived-root ratio and root weight per plant were decreased in the level of 2.0%. Survived-root ratio of seedling in sulfur solution of 1.0% was distinctly increased by 4.7 times compare to the control, but control value for root rot was relatively low as 49.2%. Mycelium growth of C. destructans, F. solani, and R. solani were distinctly inhibited by the increase of sulfur's concentration in vitro culture using PDA medium.

: This study was conducted to investigate the quality of seeds, the germination rates and the days required for germination, to examine the patterns of protein expressions during the germination and to improve the techniques of managing and storing seeds and viability of the seeds of Lithospermum erythrorhizon Sieb. et Zucc. After collecting and harvesting seeds, they were classified to white and brown colors of seed coat through testing their seed size, weight, and quality. The germination rates, the days required for germination, and the protein expressions were examined with different colors of seed coats, storing temperatures and durations by treating the different plant growth regulators and primings. One hundred seed weight of white color was heavier about 1.17 g than those of brown one about 0.81 g. The germination rates in white color of seed coat was higher, 3.05~5.75%, than those in brown one. Its rates were decreased with getting longer in storage durations. There was no big differences on germination rates between storage temperatures. The plant growth regulator of GA 3 and Kinetin was affected to improve the seed germination. GA 3 increased the seed germination clearly at 25 ppm level, while kinetin increased it gradually from 25 to 100 ppm levels. In germination by seed primings, PEG6000 made higher germination rate with increasing their levels, whereas KNO 3 increased the germination until 100 mM level and then decreased it with 200 mM unlike PEG6000. The protein expressed during the seed germination were appeared more and clearer bands in the seed after germination, especially 20~30 kDa, compared to those in the seed before germination. These results showing more and clearer bands were positively related to the germination rates which were different by seed colors, storage temperatures and durations, and plant growth regulators and primings.
