Identification of high-risk patients with clear cell renal cell carcinoma based on interphase-FISH

Abstract: Background:The aim of this study was to examine the prognostic value of four significant aberrations based on our previous studies by array-CGH to develop a prognostic Fluorescence-in situ-hybridisation (FISH) assay for clear cell renal cell carcinomas (ccRCC).Methods:Fluorescence-in situ-hybridisation experiments were performed on 100 ccRCCs (52 metastasised out of 48 non-metastasised). The mean/median follow up of patients was 59/54 months. Commercially available FISH probes were used for each critical chrom… Show more

“…In the first step, our prior findings were validated in an independent cohort of fresh frozen tumors (cohort 1). Each chromosomal aberration revealed a significant association with the metastatic status at the cut‐offs determined in the development cohort with AUC values ranging from 0.680 to 0.760. The TNSA was a much stronger indicator of metastasis compared to single aberrations, with an AUC of 0.867 ( P ‐value = 1×10 −8 ) in ROC curve analysis (Supporting Information Table ).…”

“…However, in both cohorts, the genetic score was the best independent predictor of RFS in tumors of all sizes and in organ confined tumors. The total number of specific aberrations a tumor possessed, but not single alterations, reflected their aggressiveness, although it had been demonstrated earlier that gains of chromosomes 1q, 7q, and 20q as well as loss of 9p are poor prognosis markers, leading to decreased RFS, CSS, and OS . Gains of chromosomes 1q, 7q, and 20q have been linked to tumor aggressiveness in various entities .…”

“…Es zeigte sich, dass nicht die Kombination einzelner Regionen, sondern die Anzahl der Veränderungen (0 -4) pro Tumor signifikant mit dem Auftreten von Metastasen assoziiert ist, was im sog. TNSA (total number of specific aberrations) Score definiert ist [16,17]. Nach einer ersten Validierung mittels Fluoreszenz-in-situ-Hy-▶ Tab.…”
