Identification of Highly Potent α‐Glucosidase Inhibitors from Artocarpus integer and Molecular Docking Studies

Abstract: A new natural Diels-Alder adduct (3) was isolated from the leaves and stem bark of Artocarpus integer, along with seventeen known compounds (1, 2, and 4-18). Structural elucidation was conducted using NMR and HR-ESI-MS data, and comparisons were made with previous studies. Deoxyartonin I (3) exhibited the most potent α-glucosidase inhibition (IC 50 7.80 0.1 μM), outperforming the acarbose positive control. This was mixed-mode inhibition, as indicated by the intersect in the second quadrant of each respective… Show more

“…Quercetin (68) is a natural polyphenol obtained in more than 50 plant species, as shown in the curated dataset. In terms of α-glucosidase inhibitory potential, quercetin showed superior efficacy compared to acarbose, as demonstrated by 45 out of 65 in vitro studies [ 41 , 51 , 52 , 54 , 61 , 75 , 76 , 83 , 93 95 , 98 , 101 , 105 , 114 , 116 , 141 169 ], making it the most well-examined flavonoid against α-glucosidase. It is worth mentioning that quercetin ( 68 ) was even used as a positive control in several studies.…”

“…The smaller K i of 2 means the greater binding affinity. 32 Up to now, very few polyprenyl benzoylphloroglucinols have been reported their alpha-glucosidase. Guttiferone I (2) was reported in G. cowa by Phakhatmuen and co-workers, showing a consistent result regarding alpha-glucosidase inhibition (IC 50 13.1 mM).…”

“…Among type D-I MDAAs 92-104, the diene moiety of six compounds (92)(93)(94)(95)(96)(97) Kuwanon G (92) [1] and kuwanon H (93) [2], also known as albanin F and albanin G [3] as well as moracenin B [109] and moracenin A [112], respectively, are the first reported MDAAs by three research groups in 1980 from the same plant (M. alba). Their structures were further confirmed by partial synthesis in 1981 [5].…”
