Identification of immunoglobulin E (IgE)-binding epitopes and recombinant IgE reactivities of a latex cross-reacting Indian jujube Ziz m 1 allergen

Abstract: SummaryZiz m 1 is a major Indian jujube (Zizyphus mauritiana) allergen involved in latex-fruit syndrome, and cDNA of the allergen has been cloned, sequenced and expressed in yeast by our laboratory previously. In this study, we performed an immunoglobulin E (IgE)-binding epitope analysis of Ziz m 1 using overlapping recombinant fragments. Eight overlapping recombinant fragments were generated from the recombinant Ziz m 1 allergen. The fragments were expressed in Escherichia coli and IgE-binding activities were… Show more

“…Unlike our patient, all cases reported thus far have occurred in patients who have a concomitant latex allergy [1][2][3][4]. Approximately 30-50% of patients who are allergic to latex have evidence of a coexisting food allergy, which is defined as latex-fruit syndrome [7].…”

“…One patient reacted with worsening of their atopic dermatitis and the other patient developed recurrent angioedema of the lips, tongue and throat [2]. Same authors described a case series of ten latex allergic patients who developed symptoms of upper (allergic rhinitis, oral allergy syndrome) and lower respiratory tract (asthma) after consumption of this fruit [4]. Similar to our case report, a case of systemic anaphylaxis to ingestion of five Indian jujubes in a 28-year-old female has been described.…”

“…29 Other studies indicate that a relationship between IgE clonality and the severity of allergic reactions in peanuts allergy exists. 37,38 In addition, a link between individual IgE specificities and food-induced allergic reactions [39][40][41] and a connection between IgE affinity and the outcomes of skin prick tests 42 have been found. These observations suggest that a further dissection of the IgE repertoire of the individual patient might help to relate IgE levels to disease severity.…”

“…Two stretches of residues from Asn72 - Glu86 and from Val292 - Pro320 are the possible IgE binding epitopes characterized in Ziz m 1 [142]. Recent reports suggest that this chitinase can stimulate multiple cytokines, mainly interleukin-13, from peripheral blood mononuclear cells of latex fruit allergic patients [143].…”
