“…The DNA sequence of the male characteristic fragment Md6 has been identified in cultured Pacific bluefin tuna and a BLAST search against the Pacific bluefin tuna genome assembly showed highest identity (Expect value: 8 e−139 ) with contig BADN01109032 on scaffold Ba00007445 [24]. We designed four appropriate PCR primer sets for four MS regions on the scaffold to locate Md6 on the genetic linkage map ( Supplementary Table S1).…”

“…There are multiple sex-determining regions in teleost fishes that do not possess homology to one another due to the turnover of sex chromosomes [23]. In bluefin tuna the sex-determining region has been identified to by XY based, and male-specific marker (male delta 6 or Md6) has been characterized [24]. There is no syntenic information about Md6 and those sex-determining regions in teleost fishes.…”

“…One might think this is relatively large number. Our previous study showed that there were 7 haplotypes in F3 PBF microsatellite Ttho-34, and 5 maternal line (Takagi et al 1999 andAgawa et al 2015). It was expectable that 11 polymorphism can exist in sequence level.…”

“…It is a useful strategy for aquaculture fish genotyping research. Recently, a genetic linkage map of PBF was (Uchino, Nakamura, Sekino, Kai, Fujiwara, Yasuike, Sugaya, Fukuda, Sano & Sakamoto 2016), and male sex-linked sequence (Agawa et al 2015) was mapped on it. Currently, the fragment identified in the present study has not been linked on the PBF linkage map yet, which we plan to determine in the future.…”
