Identification of Mamu-DPA1, Mamu-DQA1, and Mamu-DRA alleles in a cohort of Chinese rhesus macaques

Abstract: Rhesus macaques have long been used as animal models for various human diseases; the susceptibility and/or resistance to some of these diseases are related to the major histocompatibility complex (MHC). To gain insight into the MHC background and to facilitate the experimental use of Chinese rhesus macaques, Mamu-DPA1, Mamu-DQA1, and Mamu-DRA alleles were investigated in 30 Chinese rhesus macaques by gene cloning and sequencing. A total of 14 Mamu-DPA1, 17 Mamu-DQA1, and 9 Mamu-DRA alleles were identified in t… Show more

“…DQ genes of MHC class II region encode for α (DQA) and β (DQB) chains of the molecule (NIRANJAN et al, 2010). The second exon has been shown to be highly polymorphic and under positive selection, and the class II DQA gene has recently attracted more attention (AMILLS et al, 2008;JUNYING et al, 2008;MIYASAKA et al, 2011;HOU et al, 2011;DENG et al, 2013).…”

“…The orthologs of the human class II genes are present in macaques and are designated, respectively, Mamu-, Mafaand Mane-DR, -DQ, and -DP. Sequencing analyses have resulted in the description of about 450 class II alleles that are archived in the nonhuman primate section of the IPD-MHC database (Blancher et al 2014;Creager et al 2011;de Groot et al 2004;de Groot et al 2012;Deng et al 2013;Karl et al 2014;Otting et al 2012). However, most of these studies focused only on exon 2 sequences, which encode the antigen-binding region of the proteins (Doxiadis et al 2006;Ling et al 2011;Sano et al 2006).…”

“…The MHC-DRA gene is considered to be conserved in mammalian species, such as the DRA gene across the cattle species [ 5 , 6 ]. However, the gene has been cloned and sequenced in many other species with some variations reported: Chinese rhesus macaques [ 7 ], donkeys [ 8 ], equine [ 9 ] and sheep [ 10 ] in which allelic diversity of DRA is found to be higher than that reported in other vertebrates. Currently, 13 DRA alleles of swine have been recorded in the Immune Polymorphism Database (IPD)-SLA ( ), while only 6 DRA alleles of cattle and 11 DRA alleles of ovine are recorded.…”
