Identification of Novel Human Cdt1-binding Proteins by a Proteomics Approach: Proteolytic Regulation by APC/CCdh1

Abstract: In mammalian cells, Cdt1 activity is strictly controlled by multiple independent mechanisms, implying that it is central to the regulation of DNA replication during the cell cycle. In fact, unscheduled Cdt1 hyperfunction results in rereplication and/or chromosomal damage. Thus, it is important to understand its function and regulations precisely. We sought to comprehensively identify human Cdt1-binding proteins by a combination of Cdt1 affinity chromatography and liquid chromatography and tandem mass spectrome… Show more

“…Overexpression of Cdh1, which activates APC Cdh1 , downregulated WT Cdt1 expression ( Supplementary Fig. S5H), as previously reported 17 . Cdh1 overexpression had a comparable effect also on DPEST Cdt1, indicating that the PEST domain does not influence APC Cdh1 -mediated degradation.…”

“…A recent proteomic analysis revealed an unanticipated role of the E3 ligase APC Cdh1 in restraining Cdt1 activity 17 . D-boxes (the degrons recognized by this ligase) 40 in Cdt1 render the protein susceptible to Cdh1-mediated degradation.…”

“…4B). Its turnover is regulated by Cdh1-APCmediated degradation (31). At S phase onset, Cdt1 is degraded more efficiently by the activation of the ubiquitin ligase activity mediated by Cul4-DDB1-Cdt2 (32), and its level decreases dramatically; only low levels of Cdt1 remain.…”

“…In addition to the Skp2 pathway, PCNA/ DDB1/Cul4-dependent signaling was found to degrade Cdt1 during S phase via the interaction of Cdt1 with PCNA (11)(12)(13)(14)(15). Recently, APC/C Cdh1 was proposed as a third ubiquitin ligase regulating Cdt1 degradation (16). Cdt1 is also targeted for degradation after DNA damage to stop licensing of new origins until after DNA repair.…”
