Identification of novel sRNAs involved in biofilm formation, motility and fimbriae formation in Escherichia coli

Abstract: Bacterial small RNAs (sRNAs) are known regulators in many physiological processes. In Escherichia coli, a large number of sRNAs have been predicted, among which only about a hundred are experimentally validated. Despite considerable research, the majority of their functions remain uncovered. Therefore, collective analysis of the roles of sRNAs in specific cellular processes may provide an effective approach to identify their functions. Here, we constructed a collection of plasmids overexpressing 99 individual … Show more

“…In both LB and YESCA, the deletion of hfq had the greatest effect, essentially abolishing biofilm formation (Fig. C and D), consistent with a previous study carried out in LB (Bak et al ., ). Most of the other deletions had only modest effects, although deletion of mcaS reduced biofilm almost two‐fold in LB, and deletion of arcZ , dsrA or gadY all reduced biofilm at least two‐fold in YESCA.…”

“…Fimbrial expression can be regulated post-transcriptionally. E. coli small RNA (sRNA) have been shown to regulate the transcription of some fimbriae (265, 266). However, it is not known whether sRNA also regulate ETEC fimbriae.…”

“…46 The DNA templates for S100A11_UTR, S100A11_ORF and BC200 RNA were amplified using PCR with the corresponding primer pairs. RNAs were prepared by in vitro transcription using T7 RNA polymerase (Promega, P1320), with the amplified DNA fragments as templates.…”
