Identification of proline, 1‐pyrroline‐5‐carboxylate and glutamic acid as biomarkers of depression reflecting brain metabolism using carboxylomics, a new metabolomics method

Abstract: Aim: Depression is a psychiatric disease which is accompanied by metabolic disorder. Though depression has been widely studied, its metabolism is yet to be illustrated. We aimed to manifest the underlying mechanisms to diagnose depression. Methods: One hundred thirty serum samples, including 65 patients and 65 healthy controls from different hospitals (training and validation cohorts), were recruited into the research. Sensitive Profiling for ChemoSelective Derivatization Carboxylomics (SPCSDCarboxyl) was appli… Show more

“…In another recent report analyzing individuals of both genders (21-61 years of age), serum proline levels were up-regulated in the group diagnosed with depression, when compared to the healthy controls. This study also demonstrated higher proline levels in rats submitted to chronic mild unpredictable stress [47]. When the urinary concentrations of proline and hydroxyproline were analyzed in patients diagnosed with depression, anxiety, and stress (mean age of 34 years, both genders), the results showed that anxiety was positively correlated to proline, while hydroxyproline was weakly associated with stress.…”

“…Significant differences in glycine, L-threonine, and D-methionine between late-life depression patients and controls were revealed by this method [14]. Sensitive Profiling ChemoSelective Derivatization Carboxylomics (SPCSDCarboxyl) was proposed by Zhou's group in 2023 for the analysis of carboxylic acids using 5-(diisopropylamino) amylamine derivatization and ultra-performance LC-quadrupole time-of-flight MS (UHPLC-Q-TOF/MS) [15]. Two hundred and eight metabolites were identified in the serum of depressed patients using SPCSDCarboxyl, and a combination of proline, 1-pyrroline-5carboxylate, and glutamic acid could distinguish between patients and healthy controls.…”
