Identification of DJ‐1 as a contributor to multidrug resistance in human small‐cell lung cancer using proteomic analysis

Abstract: Proteomic approaches have been proven to provide an important tool in identifying drug resistance-associated proteins. The aim of this study was to investigate the protein profiling of drug resistance-related proteins in small-cell lung cancer (SCLC) by proteomic analysis. The proteomic profiling was performed by two-dimensional fluorescence difference gel electrophoresis (2D-DIGE) coupled with MALDI-TOF-TOF of SCLC in the multidrug-resistant cell line H69AR and its parental cell line H69. A total of 11 protei… Show more

“…PARK7 is highly expressed in patients with NSCLC and has been found to be significantly correlated with the cisplatin resistance and poor survival of patients with NSCLC [105]. PARK7 is associated with the poor survival of patients with SCLC, and markedly decreases the apoptosis of SCLC cells even after treatment with chemotherapeutic drugs, including adriamycin, cisplatin, and etoposide [106]. Proteomic analyses and bioinformatics identified PARK7 as a cisplatin-regulated gene.…”

“…Small cell lung cancer (SCLC) accounts for approximately 15% of the total cases of lung cancer (1). Compared with other types of lung cancer, SCLC is usually more sensitive to radiotherapy and chemotherapy, but its diagnosis and treatment are more difficult (3). Although SCLC is sensitive to chemotherapy, it is more likely to develop drug resistance and is highly prone to systemic metastasis (4).…”

“…Proteomic methods have been used widely to identify protein network alterations that may be linked to drugs used to treat lung cancer in terms of sensitivity and resistance [34,35,36,37]. Specifically, two-dimensional gel electrophoresis has been employed to analyze protein expression profiles and identify novel diagnostic, prognostic or predictive biomarkers in these tumors [38,39,40,41,42,43,44]. Due to the complex interactome of HSP90 and the enormous number of cellular processes in which it is involved, the use of proteomic tools for the study of this chaperone and its clients is a common approach [17,45,46,47].…”
