Identification of Staphylococcus species isolated from preputium of Aceh cattle based on 16S rRNA gene sequences analysis

Abstract: Aim:This research aimed to identify Staphylococcus species isolated from preputial swabs of healthy Aceh cattle, based on 16S ribosomal RNA gene analysis.Materials and Methods:The bacterium was isolated from preputial swabs of healthy Aceh cattle. The total DNA from the isolated bacteria was extracted using the Genomic DNA Mini Kit followed by polymerase chain reaction (PCR) amplification of the 16S rRNA gene. The product of PCR amplification was then sequenced and aligned to the known sequences in the GenBank… Show more

“…Infectious disturbance may affect the reproductive performance of the cattle's. In support of this hypothesis, various authors explained that due to the ubiquitousness of the bacteria, there are multiple potential sources of preparation infection [11], [12], [13], [14]. In confirmation of our previous study of Escherichia fergusonii [13] and Staphylococcus species [14], both bacteria were found in the preputial swabs of Aceh cattle.…”

“…Later, culture-dependent studies were conducted to isolate the bacteria responsible for bull infertility [42] and this technique was also used in more recent studies of the microbiological evaluation of frozen semen samples [43]. During the past few years, the identification of individual bacteria from preputial mucosa [44,45] and bull semen [46] was performed with metagenomic methods. Interestingly, there were no significant correlations between the preputial microbial community of bulls of different ages, breed, diet or co-housing [47].…”

“…These isolates were collected in 2019 from nasal swabs of swine in Henan province/China and were cultured on Staphylococcus chromogenic medium. After incubation at 37°C for 24–36 h, colonies were selected for 16S rRNA sequencing as described previously ( 32 ). Staphylococcus aureus RN4220 served as recipient strain in electrotransformation experiments.…”
