Identification of the caveolae/raft-mediated endocytosis as the primary entry pathway for aquareovirus

Abstract: Grass carp reovirus (GCRV), a member of the Aquareovirus genus in the Reoviridae family, is considered the most pathogenic aquareovirus. However, its productive viral entry pathways remain largely unclear. Using a combination of quantum dot (QD)-based live-virus tracking and biochemical assays, we found that extraction of cellular membrane cholesterol with methyl-β-cyclodextrin (MβCD) and nystatin strongly inhibited the internalization of GCRVs, and supplementation with cholesterol restored viral infection. In… Show more

“…This pathway is utilized by growth hormones, extracellular fluid, glycosylphosphatidylinositol (GPI)-linked proteins and interleukin-2 to enter the cells. For instance, folic acid that employs clathrin- and caveolae-independent pathway to enter the cells [ 58 , 72 , 75 79 ] are conjugated to NPs and polymers used in drug delivery systems and as imaging agents [ 53 , 80 , 81 ]. Macropinocytosis is a type of pinocytosis mechanism in which cells take in high volumes of extracellular fluid by forming a large vesicle (0.5–10 μm) called macropinosomes [ 82 85 ].…”

“…In the past decades, it has been proved that many viruses can be internalized via clathrin‐ and caveolae‐mediated endocytosis by different methods. By using specific inhibitors, RNA interference, and dominant negative mutants, it has been indicated that hepatitis C virus, African swine fever virus, hepatitis E virus, and porcine hemagglutinating encephalomyelitis virus enter their host cells via clathrin‐mediated endocytosis, while grass carp reovirus and porcine sapelovirus enter their host cells via caveolae‐mediated endocytosis . In addition to these conventional methods, single‐virus tracking provides many new insights into the study of the virus endocytic mechanism.…”

“…GCRV-I is the most widely studied type among the three types of GCRV. It is reported that caveolae/raft-mediated endocytosis is the primary entry pathway for GCRV-I [38] and disruption of clathrin-dependent trafficking results in failure of GCRV-I cellular entry [39]. Thus, the invasion mechanism of GCRV-I remains controversial.…”
