Identification of two small regulatory RNAs linked to virulence in Brucella abortus 2308

Abstract: Summary Hfq is an RNA‐binding protein that functions in post‐transcriptional gene regulation by mediating interactions between mRNAs and small regulatory RNAs (sRNAs). Two proteins encoded by BAB1_1794 and BAB2_0612 are highly over‐produced in a Brucella abortus hfq mutant compared with the parental strain, and recently, expression of orthologues of these proteins in Agrobacterium tumefaciens was shown to be regulated by two sRNAs, called AbcR1 and AbcR2. Orthologous sRNAs (likewise designated AbcR1 and AbcR2)… Show more

“…These sRNAs include AbcR1 and AbcR2 (39), orthologues of SmrC15 and SmrC16 of Rhizobium etli, and AbcR1 and AbcR2 of Agrobacterium tumefaciens. These two sRNAs are required for full virulence of B. abortus 2308.…”

“…This supports the finding that B. abortus VtlR transcription regulator target mRNAs (bab1-0310, bab1-0313, bab1-0314, and bab1-1794) are regulated by AbcR sRNAs and show higher regulatory gene expression with ∆AbcR sRNAs (38). In addition, AbcR sRNAs were reported to control the expression of Lrp-encoded gene BAB1-1587 in B. abortus, and expression decreased with ∆AbcR sRNAs; this finding highlights the importance of AbcR sRNA in the regulation of metabolism and amino acid transport (10). In contrast to the conserved region of AbcR1 sRNA, the regulation of target mRNA fusion was dependent on the seed pairing of AbcR1 sRNA.…”

“…The detection of RNA transcripts was performed using a Northern blotting protocol outlined by Caswell et al (67). Briefly, RNA from cultures grown for 3 h was isolated using an RNeasy kit (Qiagen), with DNA removed using a Turbo DNA-free kit (Ambion).…”
