Identification of Urinary microRNA Profiles in Rats That May Diagnose Hepatotoxicity

Abstract: Circulating microRNAs (miRNAs) have emerged as novel noninvasive biomarkers for several diseases and other types of tissue injury. This study tested the hypothesis that changes in the levels of urinary miRNAs correlate with liver injury induced by hepatotoxicants. Sprague-Dawley rats were administered acetaminophen (APAP) or carbon tetrachloride (CCl(4)) and one nonhepatotoxicant (penicillin/PCN). Urine samples were collected over a 24 h period after a single oral dose of APAP (1250 mg/kg), CCl(4) (2000 mg/kg)… Show more

“…While urine is beginning to be more widely used (Yang et al, 2012; Kanki et al, 2014; Yang et al, 2015), field-collected samples are still rarely measured. In this study, we were able to successfully isolate and quantify miRNA expression in field collected urine samples that had be archived in our biorepository.…”

“…The miRNeasy Serum/Plasma Kit (Qiagen) was used to purify total miRNAs in cell medium, with the C. elegans miR-39 (cel-miR-39) as the Spike-in control, according to the manufacturer’s protocol. Total RNA in serum was extracted using TRIzol (Invitrogen, Carlsbad, CA) methods as described previously (Yang et al 2012). …”

“…3A). Interestingly, miR-21 and miR-29b are reported circulatory miRNAs able to integrate into surrounding cells acting as paracrine signaling molecules (14,(18)(19)(20)(21)(22). In addition, it was recently discovered that miR-21 is able to bind to TLR8 located within endosomes of immune cells to induce an inflammatory response (16).…”
