Studies of the binding characteristics for a number of choline analogues have revealed that antiphosphorylcholine (PC) 1 antibodies from 15 different inbred mouse strains which differ in histocompatibility and heavy-chain allotype are remarkably restricted and show the same binding specificity as a PC-binding myeloma protein, HOPC 8. 2' 3 This suggests that the anti-PC antibodies of all mice possess similar binding sites for PC. Indeed Cosenza and KShler (5) and others (2, 6, 7) using mouse antimouse idiotypic antisera have dearly demonstrated that anti-PC antibody from BALB/c possesses antigenic determinants shared by HOPC 8. Paradoxically however, antibodies from other strains, such as C57BL/6, A, and CE mice (2, 5-7), do not have this idiotypic specificity even though their antibody specificity cannot be distinguished from that of BALB/c. These studies indicate that variable-region determinants differ in the anti-PC antibodies of different strains of mice, but since the hapten inhibition of binding to idiotype was not studied, it is not dear that these idiotypic differences lie in the binding region. This consideration is crucial to the analysis of antibody combining-site diversity since selective pressures exerted on binding site and nonbinding site regions may be different.

The class, subclass and light chain distribution of eight myeloma proteins with anti-streptolysin·activity (ASO) in sera studied by our group and in cases from the literature indicates a predominance of the IgG₁ kappa subclass and type of immunoglobulins. Three IgG myeloma proteins (O.I., L.F. and A.S.) were further characterized with respect to idiotypic cross-reaction and variable region subgroup. The anti-idiotypic antibodies (anti-Id) against protein O.I. and against protein L.F. both indicated idiotypic cross-reaction between O.I. and L.F. While in the case of anti-Id_L.F. this cross-reaction was in the order of 5% it was much weaker when studied with anti-Id_O.I.·The reaction between O.I. and anti-Id_O.I. could be inhibited to 95% by the antigen streptolysin O, indicating that the idiotypic determinants are associated with the combining site. N-terminal amino acid sequence analysis showed that all three light chains fall into the VκIII variable region subgroup. The heavy chains of proteins O.I. and L.F. with idiotypic similarities both have blocked N-termini, indicating the presence of the V_HI or V_HII subgroup while protein A.S. belongs to the V_HIII subgroup.

The immune response of nine inbred and one outbred strain of mice to the streptococcal group A polysaccharide was investigated with respect to magnitude and restriction. Analytical isoelectric focusing served as a tool to estimate the degree of restriction of Group A polysaccharide-specific antibodies. It proved feasible to distinguish low and intermediate from high responder strains, and to delineate strain-specificity of isoelectric focusing spectra of the immune sera. For example, immune sera of BALB/c mice, restricted high responders, and of C57BL/6 mice, heterogeneous low responders, had distinct focusing properties. Responsiveness was a dominant autosomal genetic trait in C57BL/6 x BALB/c F1 hybrid mice, irrespective of the maternal and the paternal genotype; the immune sera of these mice had their own, rather uniform isoelectric focusing spectra whereby structural genes of the low responder strain were expressed to predominant levels in 81% of the hybrids. Responsiveness in C57BL/6 x BALB/c F2 progeny segregated into 79% high and 21% low responders, and showed no genetic linkage to the following characteristics: hair color, sex, H-2 type, and Ig allotype of the heavy chain. The isoelectric focusing properties of these immune sera indicated segregation into patterns like BALB/c mice (40%), F1 hybrids (48%), and C57BL/6 mice (12%). Since this segregation is independent of any of the above criteria in these F2 mice a regulatory gene(s) is postulated that controls the clonal pattern of the immune response.
