Idiotypic Specificity of Rabbit Antibodies to Streptococcal Group Polysaccharides

Abstract: Myeloma proteins are known to possess individual and idiotypic antigenic specificlties (1-5), revealed by antisera raised within the same or a different species. Such specificifies have also been demonstrated on antibody molecules directed against various antigens (6-10). They are located in the Fab fragment of the antibody (6,(8)(9)(10). Since the individual antigenic specificity (IAS) 1 and idiotypic specificity (ID), at least in the early experiments (2, 6-8), were shown to be strictly individual and not sh… Show more

“…This association was observed regardless of strain or antibody titer and occurred throughout a primary and secondary response. This contrasts with other systems, including that defined with the mouse antisera in the present work, in which intra-and interstrain variabilities in expression of an idiotype are usual (18)(19)(20)(21)(22)(23). KShler has recent evidence in the in vitro primary anti-PC response which supports the findings presented here.…”

“…5 The fact that the H88 idiotype is present on anti-PC antibodies of all mice, and thus can be considered a species-specific idiotype, clearly distinguishes the immune response to PC from other antibody responses. In systems such as the antistreptococcal and anti-p-azohapten antibody response idiotypic cross reactions are not usually observed except among selectively bred rabbits (18,22,24) or within an inbred strain of mice (20,21,23,25). Nisonoff and colleagues (21), however, have described strong idiotypic cross-reactions among anti-pazobenzoate antibodies from BALB/c, A, and C57BL/6 mice, though the relationship of these idiotypes to combining sites was not discussed.…”

“…Idiotypic cross-reactions between antibodies of the same specificity have repeatedly been observed in inbred strains of mice [5,31] and in part-inbred rab bits [2,9]. This phenomenon, however, is not restrict ed to inbred animals, but occurs also between differ ent strains of mice [4,6] and even between different species [13,25].…”

“…It is believed that IEF and idiotypy score for different characteristics of the variable region (27). Since A-CHO antibodies of BALB/c mice were reported to belong predominantly to the IgG2, subclass (5), differences in the isoelectric points of monoclonal antibodies within the investigated pH range should not be due to substitutions of charged amino acids in the constant region of the heavy chain.…”
