IL-1 regulates in vivo C—X—C chemokine induction and neutrophil sequestration following endotoxemia

Abstract: The influx of neutrophils into tissues in response to inflammatory stimuli involves C—X—C chemokines. Interleukin-1 (IL-1) stimulates chemokine production in vitro , but its role in vivo on chemokine production is not as clearly understood. We hypothesized that IL-1 mediates in vivo tissue C—X—C chemokine production induced by systemic lipopolysaccharide (LPS). IL-1 activity was blockedbyIL-1 receptor antagonist (IL-1Ra). Rats were injected with Salmonella typhi LPS (0.5 mg/kg) with and without prior administr… Show more

“…Although IL-17 does not directly act on neutrophils, it enhances secretion of neutrophil-attracting chemokines, such as CXCL1 and CXCL8, in neighboring cells, in part by prolonging the half-life of their mRNA (58,59). Likewise, both IL-1a and IL-1b are strong inducers of CXCL1 (31,60,61). Accordingly, attenuation of IL1b secretion by IFN-g impaired CXCL1 production in target cells.…”

“…However, blocking IL-1 activity is not without its drawbacks. For example, treatment with anakinra, anti-IL-1b (canakinumab) or monoclonal anti-IL-1R1 Abs can result in neutropenia (16) and an increased risk of infections (17)(18)(19). Anakinra is contraindicated in immune-compromised patients and in patients with a history of recurrent infections.…”

“…How IL-1 in normal infected mice causes PMN influx remains unclear. However, because IL-1 itself is not chemotactic for PMN (31), the effect must be indirect, with IL-1 inducing one or more chemokines. A likely candidate is MIP-2, because MIP-2-neutralized mice develop minimal PMN responses to ocular HSV infection (30).…”
