IL-10 can induce the expression of EBV-encoded latent membrane protein-1 (LMP-1) in the absence of EBNA-2 in B lymphocytes and in Burkitt lymphoma- and NK lymphoma-derived cell lines

Abstract: IntroductionThe human gammaherpesvirus Epstein-Barr virus (EBV) is ubiquitous and it persists for the lifetime of the individual after the first encounter. In spite of the efficient transforming potential of the virus for B lymphocytes in vitro, the infection with EBV is largely harmless in vivo due to the vigorous immune response directed against the virus-encoded proteins expressed in proliferating cells, 1 combined with the viral strategy to downregulate the expression of the immunogenic viral proteins in t… Show more

“…We showed previously that IL-10 could induce the expression of LMP-1 in the absence of EBNA-2 in type I BL and NK lymphoma cell lines (23). Because both IL-10 and IL-21 potently activated STAT3, the LMP-1-inducing effect of the two cytokines probably converges on STAT3.…”

“…18 The following BL lines were studied: the type I Mutu I cl.216, cl.59, cl.148, 19 Jijoye M13; the type III Mutu III cl.99 and cl.176, 19 Raji, Jijoye P79; BLs with an EBNA-2-deleted EBV strain Daudi and P3HR1. 20 The KMH2 cells infected with the Akata-Neo strain (KMH2-EBV), made in our laboratory, were previously reported. 17 A similar pair of EBV-negative and -positive KMH2 cells (KMH2(PM) and KMH2-EBV(PM) in this study) were kindly provided by Karl R. N. Baumforth and Paul G. Murray 21 (CRUK Institute for Cancer Studies, University of Birmingham).…”

“…It has been implicated in the pathogenesis and tumor progression in several human malignancies, including Burkitt's and Hodgkin's lymphomas, gastric carcinoma and nasopharyngeal carcinoma (NPC) (Lindahl et al, 1974;Kis et al, 2006;Queiroga et al, 2008). EBV infection is mainly characterized by the expression of latent genes including nuclear antigens (EBNAs), latent membrane proteins (LMPs) and EBV-encoded RNA (EBER) (Munz and Moormann, 2008;Thorley-Lawson and Allday, 2008) under strict, host-cell-dependent transcriptional control.…”
