“…This is consistent with previous report showing that IFN-␥ is involved in differential isotype secretion, since it enhances IgG2a while suppressing IgG1 production (27). In addition, coadministration of antigens and IL-12 to mice induced a Th1-type immune response through elevation of IFN-␥ levels and consequently increasing Ag-specific IgG2a titers (17,54). The high IFN-␥ levels found in Ribiϩ6AgϩIFN-␥-immunized mice are therefore responsible for the antibody isotype switching toward the Th1-type IgG2a isotype.…”

“…Levels of IL-4 in all groups from both trials were less than 10 pg/ml (the minimum limit of detection using this kit is 10 pg/ml). This is consistent with earlier studies with TPI and the 23-kDa integral membrane protein candidate vaccine antigens driving Th1-type but not Th2-type immune responses (9,10). Levels of IL-2 and IFN-␥ for each of the two experiments are presented in Tables 3 and 4, respectively.…”

“…Several groups have suggested that cell-mediated immunity involving IFN-␥-activated cells may be one mechanism of protective immunity in the mouse schistosome challenge model (4,9,21). In this regard, in vitro studies have demonstrated that schistosomula, the presumed target stage for both SjC23 and SjCTPI vaccines, were killed by macrophages or endothelial cells activated via combinations of cytokines, including IFN-␥ and IL-12 (33).…”

“…In addition, using DNA encoding S. mansoni 28 kDa glutathione S-transferase to immunize rats leads to a long-lasting predominance of IgG2a and IgG2b (Dupre et al 1997), as does the p80 subunit of S. mansoni calpain (Siddiqui et al 2003). DNA vaccination and co-delivery of antigens with IL-12 result in amplified IgG2a synthesis and down-regulation of synthesis of IgG1 (Fonseca et al 2004, Siddiqui et al 2003. Interestingly, our study demonstrated that vaccination with rSVLBP induced a predominantly a Th1-type of immune response in mice.…”
