IL15RA Drives Antagonistic Mechanisms of Cancer Development and Immune Control in Lymphocyte-Enriched Triple-Negative Breast Cancers

Abstract: Despite its aggressive nature, triple-negative breast cancer (TNBC) often exhibits leucocyte infiltrations that correlate with favorable prognosis. In this study, we offer an explanation for this apparent conundrum by defining TNBC cell subsets that overexpress the IL15 immune receptor IL15RA. This receptor usually forms a heterotrimer with the IL2 receptors IL2RB and IL2RG, which regulates the proliferation and differentiation of cytotoxic T cells and NK cells. However, unlike IL15RA, the IL2RB and IL2RG rece… Show more

“…Recently, IL‐15 has been shown to signal via IL‐15Rα in the absence of IL‐15Rβ or IL‐15Rγc . However, IL‐15Rβ‐ and IL‐15Rγc‐independent signaling did not activate STAT5 but rather the protein kinase B pathway suggesting an alternative mechanism for the reduced IL‐15 responsiveness of TRAF2TKO cells in vitro.…”

“…Interestingly, GSEA revealed an IRF1-dependent and IFN-γ-induced shift towards a Th1/IFN-like phenotype, while suppressing Th9 signature genes. Pathways associated with infectious and interferon response36505156, for example, Gbp and Il2rb1 were directly upregulated, indicating that IRF1 balances Th9 versus Th1-like plasticity by its ability to act as a transcriptional repressor as well as an activator. Recently, Végran et al 9.…”

“…BMDMs stimulated with IFN express IL-15R (42), which can transduce IL-15 signals in the absence of CD122 in certain cell types (43). IL-15-deficient mice do not remodel uterine spiral arteries properly, which is a pathologic hallmark of human preeclampsia and has been attributed to impaired production of IFN by NK cells (14).…”
