Iloperidone in the treatment of schizophrenia: an evidence-based review of its place in therapy

Abstract: IntroductionSchizophrenia is a chronic and debilitating mental disorder that affects the patient’s and their family’s quality of life, as well as financial costs and health care settings. Despite the variety of available antipsychotics, optimal treatment outcomes are not always achieved. Novel drugs, such as iloperidone, can provide more effective, tolerable and safer strategies.AimTo review the evidence for the clinical impact of iloperidone on the treatment of patients with schizophrenia.Evidence reviewClini… Show more

“…Iloperidone, a AAP drug, presents binding affinity to serotoninergic (5-HT2A, 5-HT6 and 5-HT7), dopaminergic (D2, D3 and D4) and adrenergic (α1 and α2C) receptors in the CNS) [ 142 ]. Iloperidone can inhibit serotonergic 5HT2A receptor (J-13) and adrenergic alpha 1A receptor [ 143 , 144 ] and is responsible for hypotension [ 145 ]. In dose 0.03 mg/Kg i.p., Iloperidone present reduced blood pressure within the 10 min of administration for animals [ 146 ].…”

“…9 Iloperidone has a high affinity for both D2 and 5HT2A receptors. 10 Paliperidone or 9-hydroxy risperidone is the major active metabolite of risperidone. Paliperidone acts on dopamine D2 and serotonin 5HT2A receptors as an antagonist and also antagonizes the α1-and α2adrenergic receptors and H1-histaminergic receptors.…”
