Immobilization and characterization of bovine liver catalase on eggshell

Abstract: Bovine liver catalase immobilized on eggshell particles was characterized and the reusability of the immobilized catalase was investigated in a batch type reactor. For immobilized catalase onto ground eggshell (ICATG), the optimum initial amount of catalase was 85 mg g -1 of eggshells, the optimum pH was 6.0 (75 mM citrate buffer) and the temperature was 30 °C. The V max and K m values of ICATG were determined as 29.1±1.2 U/mg of protein and 41.9±2.7 mM, respectively. The reusability of ICATG was tested and th… Show more

“…1), although both types of honey had similar GOX activity (Table 2). However, what is the real effect of catalase on the H 2 O 2 decomposition, still remains to be elucidated, especially considering the effect of low pH on the significantly decreased catalase activity (Alptekin et al, 2008). Vitamin C (ascorbic acid) is an antioxidant compound whose concentration in honeys of different botanical origin ranges from zero (Gheldof et al, 2002) up to 4250 mg/kg of honey (Chis et al, 2016;Ciulu et al, 2011;Kadri et al, 2017;Kerkvliet, 1996;León-Ruiz et al, 2011;Perna et al, 2013).…”

“…The beads were kept in this solution for 5 h at 25 °C in a rotating incubator. The immobilized beads were washed with cold phosphate buffer (pH 7.5) thoroughly and stored for further study [ 4 , 16 , 20 ].…”

“…Catalase is a common enzyme found in nearly all living organisms aerobic as well as anaerobic. It has one of the highest turnover of all enzymes as it has the capacity to decompose more than one million molecules of hydrogen peroxide per molecule of enzyme [ [2] , [3] , [4] , [5] ]. Catalase is a tetramer of four polypeptide chains, each over 500 amino acids long.…”

“…Adsorption, encapsulation, entrapment and covalent binding (Kondo et al, 1993;Alptekin et al, 2008Alptekin et al, , 2009Alptekin et al, and 2010Itoh et al, 2009) have been used to immobilize CAT. However, Wilson et al (2004) reported that these methods are difficult to stabilize the CAT quaternary structure.…”
